      驼蛾总科Hyblaeoidea隶属于鳞翅目Lepidoptera螟蛾类Obtectomera,其系统发育位置一直存在争议,驼蛾幼虫取食柚木和红树植物,具有一定的危害性,其线粒体基因组数据在生态和进化研究方面具有重要意义。本研究通过高通量测序获得了白星驼蛾Hyblaea constellata Guenée, 1852线粒体全基因组序列并对其主要特征开展分析,结果表明:(1) 白星驼蛾线粒体基因组全长为15 317 bp,包含13个蛋白质编码基因,22个转运RNA基因,2个核糖体RNA基因和一个非编码控制区,其基因排列顺序与大多数已报道的鳞翅目昆虫相同,呈tRNA-Met-tRNA-Ile-tRNA-Gln排列。(2) 除COI以CGA作为起始密码子,COII和ND5以T作为不完全的终止密码子以外,其余蛋白质编码基因都以典型的ATN作为起始密码子,以典型的TAN作为终止密码子。除tRNA-Ser (GCU) 缺少DHU臂以外,其余tRNA均可形成典型的三叶草二级结构。16S rRNA的结构域IV和V比结构域I、II、VI更保守,12S rRNA的结构域III比结构域I和II更保守。(3) 核苷酸组成和密码子使用都表现出了很高的AT偏向性,使用最频繁的密码子均由AT组成且多数不与tRNA反密码子严格配对。此外,结合11总科27种已报道的鳞翅目线粒体基因组数据开展了系统发育分析,结果支持驼蛾总科与螟蛾总科+大鳞翅类互为姐妹群。

      Hyblaeoidea belongs to Obtectomera of Lepidoptera, and its phylogenetic position remains controversial. The larvae of Hyblaeoidea are important pests and feed on teak and mangrove plants. The data of mitochondrial genomes of Hyblaeoidea are playing principal roles in ecological and evolutionary research. In this study, the complete mitogenome sequence of Hyblaea constellata Guenée, 1852 is determined by high-throughput sequencing and its main characteristics are analyzed. The results show that: (1) The total length of the mitogenome of H. constellata is 15 317 bp, including 13 protein-coding genes, 22 transfer RNA genes, 2 ribosomal RNA genes and a non-coding control region. The genes organization are same as those of mostly reported Lepidoptera insects, with an order of tRNA-Met-tRNA-Ile-tRNA-Gln. (2) Except COI starting with CGA, COII and ND5 terminating with incomplete codon T, for all other protein-coding genes, the typical ATN is used as the start codon with TAN as the stop codon. All tRNA could be presented with a typical secondary structure of cloverleaf except tRNA-Ser (GCU) lacking DHU stem. Domains IV and V of 16S rRNA are more conserved than domains I, II, and VI, and domain III of 12S rRNA is more conserved than domains I and II. (3) Both nucleotide composition and codon usage show a high AT bias. The most frequently used codons are all composed of AT, and most of them are not strictly paired with tRNA anticodons.In addition, combined with the reported mitochondrial genome data of 27 species in 11 superfamilies of Lepidoptera, the phylogenetic analysis supports that Hyblaeoidea and Pyraloidea+Macrolepidoptera are sister relationship to each other.
