Fox家族蛋白广泛存在于植物以外的物种中,是一类与发育相关的重要转录因子。但是不同Fox基因在鳞翅目昆虫家蚕 Bombyx mori 精巢中的表达特征,以及BmFoxL2亚家族蛋白在精巢发育中的功能未知。本文检测了家蚕12个 BmFox 基因在家蚕精巢中的表达量,发现 BmFoxL2-2的表达量最高,BmFoxL2-1次之。BmFoxL2-1和BmFoxL2-2在精巢中的表达量显著高于卵巢。亚细胞定位分析显示BmFoxL2-1在细胞核中表达,BmFoxL2-2在细胞核和细胞质中都有表达。生物信息学分析显示BmFoxL2-1和BmFoxL2-2都有保守的Fox结构域,但N端结构以及理化性质存在差异。BmFoxL2-1在家蚕精巢中的表达随着发育时间逐渐升高,BmFoxL2-2的表达在幼虫末期和蛹期达到高峰;BmFoxL2-1和BmFoxL2-2在精巢内容物中的表达均高于精巢膜。在Bm12细胞中分别过表达BmFoxL2-1和BmFoxL2-2蛋白后,BmNanos、BmVasa和BmCyclin A等发育相关基因的表达均显著上调,暗示这些基因受BmFoxL2亚族蛋白的调控。由此,本文推测家蚕FoxL2亚族蛋白可能通过调节精巢细胞周期或生殖细胞的功能来调节家蚕精巢的发育。 |

The Forkhead box (Fox) proteins are critical transcription factors involved in the development of many organisms but plants. However, the expression patterns of Bombyx mori Fox genes in the testis and function of BmFoxL2 subfamily during testis development remain unknown. In this study, the expression patterns of 12 BmFox genes in the testis were analyzed BmFoxL2-2 had a highest expression level and followed by BmFoxL2-1 Both BmFoxL2-1 and BmFoxL2-2 were expressed at a significantly higher level in the testis than in ovary. BmFoxL2-1 was expressed only in the nucleus, and BmFoxL2-2 was located in both the nucleus and cytoplasm. Both BmFoxL2 proteins contained the conserved Forkhead domain, and they varied in N-terminus, physical and chemical characteristics. The transcript of BmFoxL2-1 gene was increased during development, BmFoxL2-2 mRNA was abundant in the testis at the late stage of both larvae and pupae; BmFoxL2 gene was expressed at a higher level in the sperm than in the testis membrane. Genes participate in development, including BmNanos, BmVasa and BmCyclinA, were upregulated in Bm12 cells overexpressing BmFoxL2-1 and BmFoxL2-2. Our results suggested that FoxL2 subfamily proteins might participate in testis development by regulating cell cycles or spermatogenesis in B.mori. |
