      为了分析番茄潜叶蛾Tuta absoluta (Meyrick)不同地理种群中肠内共生菌多样性及相对丰度,本研究通过16S rDNA高通量测序技术分析了番茄潜叶蛾中肠内共生菌多样性及相对丰度,并通过KEGG分析了内共生菌生物群落基因代谢途径差异。结果表明,酒泉种群的丰富度、多样性、均匀度和进化多样性等方面均相对高于张掖。两个地理种群丰度前20的中肠细菌分别隶属于放线菌门Acfinobacteria、拟杆菌门Bacteroidetes、厚壁菌门Firmicutes、变形菌门Proteobacteria和疣微菌门Verrucomicrobia,其中均以厚壁菌门和变形菌门细菌相对丰度较高。张掖地理种群相对丰度最高的为沃尔巴克菌属Wolbachia(16.06%),隶属于变形菌门;酒泉地理种群相对丰度最高的为乳杆菌属Lactobacillus(13.75%),隶属于厚壁菌门;两个地理种群相对丰度总和最高且差异最大的为沃尔巴克菌属,张掖地理种群的相对丰度为酒泉地理种群的18.89倍。另外,本研究对微生物群落的33种基因功能进行了预测,主要群落基因功能表现为代谢功能,其中碳水化合物代谢功能最强,且种群间没有显著差异;两个地理种群间代谢功能差异最显著的为异生物降解和代谢功能,以酒泉种群相对较强。总体而言,两个地理种群的中肠细菌多样性差异较小,其中寡养单胞菌属Stenotrophomonas细菌可能与害虫抗性密切相关,沃尔巴克菌属细菌可应用于害虫生物防治,前景良好。

      In order to analyze the diversity and relative abundance of intestinal endosymbiotic bacteria in different geographic populations of tomato leaf miner moth, this study analyzed the diversity and relative abundance of intestinal endosymbiotic bacteria in tomato leaf miner moth by 16S rDNA high-throughput sequencing, and analyzed the differences in gene metabolic pathways of endosymbiotic bacterial biomes by KEGG. The results showed that the Jiuquan population was relatively higher than Zhangye in terms of abundance, diversity, evenness and evolutionary diversity. The top 20 midgut bacteria in the abundance of the two geographic populations belonged to Actinomyceta, Bacteroides, Firmicutes, Proteobacterium and Verrucomicrobia, respectively, of which the relative abundance of Firmicutes and Proteobacteria bacteria were all higher. The highest relative abundance of Zhangye geographic population was Wolbachia (16.06%), which belongs to Proteobacteria; the highest relative abundance of Jiuquan geographic population was Lactobacillus (13.75%), which belongs to Firmicutes; and the largest difference in relative abundance between two geographic populations was in the Wolbachia genus, with the relative abundance of the Zhangye geographic population being 18.89 times higher than that of the Jiuquan geographic population. In addition, 33 gene functions of microbial communities were predicted in this study, and the main community gene functions were metabolic functions, among which carbohydrate metabolism was the strongest, and there was no significant difference between populations; the most significant differences in metabolic functions between two geographic populations were xenobiotics biodegradation and metabolism, with Jiuquan populations being relatively stronger. Overall, the diversity of bacteria in the midgut of the two geographical populations was small, among the species, Stenotrophomonas might be closely related to the resistance of pests, and the bacteria of Wolbachia could be applied to biological control of pests with a good prospect.
