      通过单因素试验和正交试验探讨从黑皱鳃金龟中提取甲壳素的最佳工艺条件。利用氢氧化钠溶液和柠檬酸溶液依次对黑皱鳃金龟干粉进行处理后,用紫外吸收法和干法灰化法分别测定提取物中残余蛋白质和灰分含量以检验脱除效果,并在最佳工艺条件下,提取甲壳素样品,对其进行定性和定量分析。试验结果表明:利用酸碱法提取甲壳素的最佳工艺条件为:氢氧化钠浓度8%、固液比1∶10、温度85℃ 、时间4 h,柠檬酸浓度1.6 g/L、时间25 h、固液比1∶12、室温,对在此工艺条件下提取得到的甲壳素样品进行红外光谱检测并计算其得率为24.82%。

      This article discussed the optimum process of extracting chitin from Trematodes tenebrioides (Pallas) by single-factor and orthogonal test. The powder of T. tenebrioides was treated by the solution of sodium hydroxide and hydrochloric acid successively. Then residual protein and inorganic salt content in the extraction were measured to examine removal effect by UV absorption and dry ashing method. The chitin samples extracted under the optimum process then carried on the qualitative and quantitative analysis. Test results showed that the optimum process by using acid-alkali method to extract chitin were as follows: the NaOH density 8%, the solid-liquid ratio 1∶10, the temperature 85℃, the time 4 h, the citric acid density 1.6 g/L, the time 25 h, the solid-liquid ratio 1∶12, at room temperature. Using infrared spectrum to analysis chitin samples extracted under optimum process and the extraction rate of chitin was 24.82%.
