      本文克隆了虫害诱导的水稻脂氧合酶基因OsRCI-1全编码序列, 实时定量QRT-PCR 检测结果表明二化螟Chilo suppressalia Walker取食能快速且持续地诱导OsRCI-1基因的表达。利用农杆菌介导的水稻遗传转化获得了两株OsRCI-1基因的RNAi沉默突变体,突变体株系ir-rci-1和ir-rci-2中目的基因OsRCI-1的表达被明显的抑制,其表达水平分别仅为对照(秀水11)的4773%和3233%。生测结果表明OsRCI-1基因沉默导致水稻对二化螟抗性显著地降低,取食RNAi突变体株系的二化螟体重分别是取食秀水11的158倍和215倍。因此,水稻OsRCI-1基因正调控对二化螟的抗性,该基因介导的防御途径可能在水稻虫害诱导防御反应中起重要的作用。

      We isolated an herbivore-induced rice lipoxygenase gene OsRCI-1 with full coding sequence (CDS) and its expression levels quickly and constantly increased after rice Striped stem borer (SSB), Chilo suppressalia Walker treatment Using Agrobacterium-mediated transformation technology, we develop two mutant lines of OsRCI-1 (ir-rci-1 and ir-rci-2),in which the expression levels of OsRCI-1 were inhibited significantly, the transcript level of OsRCI-1 in ir-rci-1 and -2 was only 4773% and 3233% of level of XS11 Bioassay showed that resistance toward SSB was greatly impaired in mutant rice plants ir-rci lines compared to wild type rice (WT). The mass of SSB that fed on ir-rci lines was 158 and 215 times comparing with WT plants, respectively The results indicated that OsRCI-1 gene positively regulated resistance of rice toward rice SSB, which may play an important role in herbivore-indcued defence in rice.
