| 【目的】核纤层是许多DNA病毒复制的天然屏障。杆状病毒是重要的昆虫病原微生物及微生物杀虫剂,其生活周期可产生两种病毒粒子:芽生型病毒粒子(Budded virus,BV)和包埋型病毒粒子(Occlusion-derived virus,ODV)。本团队前期研究已阐明核纤层解聚对BV产生的影响,但其对ODV装配的作用尚不明确。本文旨在揭示核纤层解聚对杆状病毒ODV装配的影响,以进一步阐明杆状病毒高效侵染昆虫的机制。【方法】利用透射电镜观察使用细胞周期蛋白依赖性激酶1的抑制剂抑制核纤层解聚后,杆状病毒感染的昆虫细胞中核内微囊泡形成及ODV装配的变化。【结果】抑制核纤层解聚不影响病毒发生基质中正常装配的核衣壳向环带区的转运,但显著减少核内微囊泡的形成,同时导致附着核衣壳的囊泡中间结构异常积累,最终抑制ODV的装配。【结论】核纤层是杆状病毒在昆虫细胞内产生ODV的天然物理屏障。杆状病毒通过诱导核纤层解聚突破该屏障,进而促进核内微囊泡形成及ODV装配,最终利于其在昆虫宿主中高效增殖。本文的研究为深入理解杆状病毒侵染昆虫的分子机制提供了理论依据。 |

| 【Aim】The nuclear lamina represents a natural barrier to the replication of many DNA viruses. Baculoviruses are important insect pathogens and microbial insecticides. They produce two types of virions during their life cycle: budded virus (BV) and occlusion-derived virus (ODV). Our previous study has clarified the effect of nuclear lamina disruption on BV production, but its role in ODV assembly remains unclear. This study aims to reveal the effect of nuclear lamina disruption on baculovirus ODV assembly, thereby further elucidating the mechanism underlying efficient baculovirus infection in insects.【Methods】Transmission electron microscopy was used to examine intranuclear microvesicle formation and ODV assembly in baculovirus-infected insect cells treated with a cyclin-dependent kinase 1 inhibitor to inhibit nuclear lamina disruption.【Results】Inhibition of nuclear lamina disruption did not affect the nucleocapsid transport to the ring zone, but significantly reduced intranuclear microvesicle formation and led to the abnormal accumulation of nucleocapsid-attached vesicle intermediates, ultimately resulting in the inhibition of ODV assembly.【Conclusion】The nuclear lamina serves as a natural physical barrier to ODV morphogenesis in baculovirus-infected insect cells. Baculoviruses overcome this barrier by inducing nuclear lamina disruption, thereby promoting intranuclear microvesicle formation and ODV assembly, and ultimately facilitating their efficient proliferation in insect hosts. This study provides a theoretical foundation for further understanding the molecular mechanisms of baculovirus infection in insects. |
