| 【目的】草地贪夜蛾Spodoptera frugiperda是全球性重大迁飞性农业害虫,其寄主定位、交配繁殖等关键行为的完成高度依赖嗅觉系统,而气味受体(Odorant receptor,OR)是嗅觉信号通路中的关键分子。本研究以室内连续饲养的草地贪夜蛾种群为研究对象,探究该种群OR基因转录和表达特征反映的嗅觉可塑性。【方法】结合生物信息学与分子试验手段,对其OR基因家族进行系统注释,并评价不同性别和组织间的嗅觉可塑性差异。筛选并克隆4个表达OR基因(SfruOR13、SfruOR19、SfruORco、SfruOR42),利用qRT-PCR检测其在雌雄成虫触角、喙和下唇须中的表达模式。【结果】基于基因组数据共鉴定48条全长OR基因,并通过系统发育、Motif、结构域和基因结构分析推测其分支特征及功能演化。通过qRT-PCR分析其在触角、喙和下唇须等嗅觉相关组织中的表达模式;结果显示4个OR基因在雌雄成虫间表达存在显著差异,其中雌虫表达水平普遍较高。【结论】研究表明室内种群在气味受体水平上表现出明显的性别差异性和嗅觉可塑性。本研究不仅丰富了草地贪夜蛾室内种群嗅觉分子机制的研究,也为基于OR的绿色防控技术(如高效引诱剂和驱避剂设计)提供了潜在靶标和理论支撑。 |

| 【Aim】The fall armyworm Spodoptera frugiperda is a globally significant migratory agricultural pest, whose key behaviors such as host location and mating reproduction are highly dependent on the olfactory system, with odorant receptors (ORs) serving as key molecules in the olfactory signaling pathway. This study aimed to investigate the olfactory plasticity reflected by the transcriptional and expression profiles of OR genes in a laboratory-reared S. frugiperda population. 【Methods】Using an integrated approach combining bioinformatics and molecular experiments, we systematically annotated the OR gene family and evaluated olfactory plasticity across different sexes and tissues. Four expressed OR genes (SfruOR13, SfruOR19, SfruORco, and SfruOR42) were selected, cloned, and their expression patterns in antennae, proboscis, and labial palps of male and female adults were examined using qRT-PCR. 【Results】A total of 48 full-length OR genes were identified from genomic data, and their phylogenetic relationships, motif compositions, domain architectures, and gene structures were analyzed to infer their evolutionary divergence and functional conservation. qRT-PCR revealed significant differences in the expression of the four OR genes between sexes, with generally higher expression levels in females. 【Conclusion】These findings demonstrate notable sexual dimorphism and olfactory plasticity at the odorant receptor level in the laboratory population. This study not only enhances the understanding of olfactory molecular mechanisms in S. frugiperda under laboratory conditions but also provides potential targets and a theoretical foundation for developing OR-based green control strategies, such as the design of effective attractants and repellents. |
