      【目的】SMARCE1是形成染色质重塑复合物核心的重要组成,在基因表达、细胞分裂等生物过程中发挥重要作用。为了明确SMARCE1基因在斜纹夜蛾Spodoptera litura发育过程中的功能,本研究克隆获得了斜纹夜蛾SlSMARCE1基因,该基因所编码的蛋白质含570个氨基酸残基。【方法】多重序列比对和系统发育分析表明,SlSMARCE1蛋白与其他昆虫的SMARCE1蛋白具有高度保守性和同源性。在斜纹夜蛾幼虫的生长和发育过程中,SlSMARCE1主要在3龄幼虫阶段表达,5龄幼虫不同组织中的表达结果显示,SlSMARCE1在中肠中具有高表达水平。为了验证SlSMARCE1的功能,本研究探究了斜纹夜蛾SlSMARCE1基因的特性及其在RNA干扰条件下的生物学效应。【结果】结果表明,在幼虫发育过程中,RNA干扰使SlSMARCE1沉默可显著抑制斜纹夜蛾幼虫的生长发育,导致幼虫体重下降,体型变小。此外,向1龄末幼虫喷洒dsSlSMARCE1后,幼虫在72 h内的存活率为55%,且RNAi通路的关键基因Ago与Dicer的表达显著升高。【结论】这些结果不仅表明SlSMARCE1在斜纹夜蛾的生长发育过程中具有重要的作用,同时还为利用RNAi技术防控斜纹夜蛾提供新的靶标基因,为在农业害虫防治中减少对化学杀虫剂的依赖提供了新思路。

      【Aim】SMARCE1, an essential component that forms the core of chromatin remodeling complexes, plays a significant role in various biological processes including gene expression regulation and cell division. To elucidate the functional role of SMARCE1 in the developmental process of Spodoptera litura, we cloned and characterized the SMARCE1 gene in this study. The protein it encodes consists of 570 amino acid protein residues.【Methods】Multiple sequence alignment and the phylogenetic analysis revealed that SlSMARCE1 protein exhibited a high degree of conservation and significant homology with SMARCE1 proteins in other insects. During S. litura larval development, SlSMARCE1 exhibited stage-specific expression with peak levels observed in third-instar larvae. Tissue-specific profiling in fifth-instar larvae revealed predominant SlSMARCE1 expression in the midgut. To verify the function of SlSMARCE1, this study investigated the characteristics of the gene in S. litura and its biological effects under RNA interference.【Results】The results showed that silencing of SlSMARCE1 by RNA interference significantly inhibited the growth and development, decreased larval weight and size of S. litura larvae during larval development. Furthermore, after spraying dsSlSMARCE1 to larvae at the end of the 1st instar, the survival rate of larvae was 55% within at 72 h, and the expression of Ago and Dicer, key genes of the RNAi pathway elevated significantly.【Conclusion】These results not only demonstrate that SlSMARCE1 has an important role in the growth and development of S. litura, but also provide new target genes for the prevention and control of the S. litura using RNAi technology, and provide a promising strategy for reducing the reliance on chemical insecticides in agricultural pest control.
