| 瓜实蝇Zeugodacus cucurbitae(Coquillett)是一种危害极为严重的农业检疫性害虫,抑制瓜实蝇雌虫卵巢发育及繁殖能力是遏制其种群扩张的关键因素。本研究从调控瓜实蝇雌虫卵巢发育的关键基因入手,通过雌成虫的脂肪体、中肠、表皮、马氏管、卵巢以及雄成虫等不同性别或组织的RT-qPCR验证,筛选鉴定出3个与雌性生殖相关的关键基因:卵巢高表达基因chorion protein s36 transcript variant X3(s36),以及卵巢特异性表达基因pickpocket protein 28(pp28)、fascetto(feo),并对其在瓜实蝇发育历期中的表达模式进行解析,随后利用RNAi技术阐明了s36、pp28、feo在雌性瓜实蝇繁殖中的调控功能。结果显示,逐一沉默3日龄雌成虫的s36、pp28、feo后,瓜实蝇雌虫的死亡率均显著增加,卵巢发育受到明显抑制,产卵量及孵化率显著降低,同时卵黄原蛋白(Vg)的表达也出现显著下调。本研究表明,卵巢特异性基因pp28、feo和高表达基因s36对瓜实蝇雌虫的繁殖和卵巢发育具有重要调控作用,为瓜实蝇绿色防控提供了有效靶标。 |

| Zeugodacus cucurbitae (Coquillett) (Diptera: Tephritidae) is a highly destructive agricultural quarantine pest, and inhibiting its ovarian development and reproductive capacity represents a key strategy for controlling population expansion.This study focused on key genes regulating ovarian development in female Z. cucurbitae. Through RT-qPCR validation across different tissues—including fat body, midgut, epidermis, Malpighian tubules, ovaries of females, and both sexes (female/male adults)—three key female-reproduction-related genes were identified: the ovary highly expressed gene chorion protein s36 transcript variant X3 (s36), and the ovary-specific genes pickpocket protein 28 (pp28) and fascetto (feo). Their expression patterns throughout the developmental stages of Z. cucurbitae were analyzed. Subsequently, RNA interference (RNAi) was employed to investigate the regulatory roles of s36, pp28, and feo in female reproduction. The results showed that individually silencing s36, pp28, or feo in 3-day-old female adults significantly increased mortality, suppressed ovarian development, and markedly reduced egg production and hatchability. Additionally, the expression of vitellogenin (Vg) was significantly downregulated. This study demonstrates that ovary-specific genes (pp28, feo) and the highly expressed gene (s36) play crucial regulatory roles in the reproduction and ovarian development of female Z. cucurbitae, providing potential molecular targets for eco-friendly pest control strategies. |
