为明确七星瓢虫Coccinella septempunctata 取食人工饲料和豆蚜Aphis craccivora的生物特性分子机制和营养代谢通路,利用高通量测序技术分析七星瓢虫4龄幼虫和成虫的相对转录水平。结果显示,从18个样本中共筛选出1 234 640 650条干净序列(clean reads),与参考基因组比对到813 335 465条序列。通过DESeq2软件分析,其中有11 120个差异基因(DEGs)表达水平发生了显著的变化,总共有2 280条DEGs被注释到KEGG通路中。在取食人工饲料的七星瓢虫中,与生物学特性相关的细胞色素P450(Cytochrome P450)、保幼激素环氧水解酶(Juvenile hormone epoxide hydrolase,JHEH)、保幼激素酯酶(Juvenile-hormone esterase, JHE)、保幼激素III合成酶(Juvenile hormone-III synthase,JHAMT)、多巴脱羧酶(Dopa decarboxylase, DDC)和几丁质酶(Chitinase)大部分DEGs在4龄幼虫、雌雄成虫中下调,精液蛋白基因α-酮戊二酸脱氢酶(2-oxoglutarate dehydrogenase,sucA)和胰岛素样生长因子2(Insulin-like growth factor 2)DEGs在雌、雄成虫中均上调,脂蛋白(Lipoprotein)DEGs在雌虫中大部分上调,蜕皮诱导蛋白(Ecdysone-induced protein)基因在幼虫中下调。DEGs在KEGG中富集到6个氨基酸、7个脂类代谢、2个淀粉和糖代谢、6个维生素相关代谢通路。其中取食人工饲料的七星瓢虫大部分氨基酸代谢通路基因下调;脂类代谢在成虫中大部分下调,而在幼虫中大部分上调;淀粉和糖代谢在成虫中大部分上调,但在幼虫中大部分下调;维生素在幼虫中大部分下调。qRT-PCR验证证明了转录组测序获得序列质量的可靠性,可作为后续试验的研究基础。这些转录组数据为进一步优化人工饲料提供了参考依据,促进人工饲料配方的改良。 |

To clarify the biological characteristics and metabolic pathways of Coccinella septempunctata feeding on artificial diets and Aphis craccivora, high-throughput sequencing technology was used to analyze relative transcription levels of C. septempunctata in 4th instar larvae and adults. A total of 1 234 640 650 clean reads had been screened out from 18 samples and 813 335 465 sequences have been compared the same with reference genome. DESeq2 analysis indicated that the expression levels of 11 120 differentially expressed genes (DEGs) were significantly changed, and a total of 2 280 DEGs were annotated in the KEGG pathway. Among C. septempunctata feeding on artificial diets, Cytochrome P450, Juvenile hormone epoxide hydrolase (JHEH), Juvenile hormone esterase (JHE), Juvenile hormone III synthase (JHAMT), Dopa decarboxylase (DDC), Chitinase and most DEGs were down-regulated in 4th instar larvae and male and female adults. DEGs of semen protein gene, 2-oxoglutarate dehydrogenase (sucA) and Insulin-like growth factor 2 upregulated in both male and female adults, and DEGs of lipoprotein upregulated in most females; Ecdysone-induced protein genes down-regulated in larvae. DEGs had enriched metabolic pathways related to 6 amino acids, 7 fat metabolisms, 2 starch and sugar metabolisms, and 7 vitamin-related metabolisms in KEGG. Among them, most genes of amino acid metabolism pathway of the C. septempunctata feeding on artificial diets down-regulated; whereas fat metabolism mostly down-regulated in adults and upregulated in larvae, starch and sugar metabolisms mostly upregulated in adults and mostly down-regulated in larvae, and vitamin mostly down-regulated in larvae. Quantitative real-time PCR verification showed that the sequence quality of transcriptome met the basic requirements for transcriptome data analysis. These transcriptome data provide reference for further optimization of artificial diet and promote the improvement of artificial diet formula. |
