      本研究旨在阐明蛋白磷酸酶2A支架亚基a(Protein phosphatase 2A subunit a,PP2Aa)在美洲大蠊Periplaneta americana断肢再生中的功能。利用生物信息学分析了PP2Aa蛋白的系统进化关系以及氨基酸序列的保守性。利用实时荧光定量PCR技术(RT-PCR)检测了PP2Aa在美洲大蠊断肢后不同时间点的表达水平,结果显示,在断肢后3~120 h内PP2Aa表达量上调,断肢240 h后PP2Aa表达量下降,暗示PP2Aa参与了美洲大蠊断肢再生过程中的基芽形成和形态建成。通过注射PP2Aa dsRNA以干扰其表达,发现抑制PP2Aa表达严重抑制了美洲大蠊的断肢再生,表明PP2Aa在美洲大蠊的断肢再生的过程中发挥重要作用。进一步利用EdU和DAPI染色技术证实干扰PP2Aa后,细胞增殖过程受到抑制,并且断肢处没有明显的再生腿出现,说明PP2Aa通过调控细胞增殖来促进断肢再生的进程。PP2A是一种广泛存在并且进化上非常保守的丝氨酸/苏氨酸磷酸酶,研究结果为进一步解析高等哺乳动物有限的再生能力奠定了基础。

      This study aimed to investigate the role of the protein phosphatase 2A regulatory subunit a (PP2Aa) in limb regeneration in Periplaneta americana. Bioinformatics analyses were conducted to assess the phylogenetic relationship and amino acid sequence conservation of the PP2Aa protein. Quantitative real-time PCR (RT-PCR) was used to measure PP2Aa expression at various time points following limb amputation. The results revealed that PP2Aa expression upregulated between 3 and 120 hours post-amputation, with a subsequent decline after 240 hours. These findings suggest that PP2Aa was involved in blastema formation and morphogenesis during limb regeneration in P. americana. Furthermore, the injection of PP2Aa dsRNA to inhibit its expression significantly impaired limb regeneration of P. americana, highlighting the critical role of PP2Aa in this process. Further studies using EdU and DAPI staining confirmed that the disruption of PP2Aa expression inhibited cell proliferation, preventing the formation of a visible regenerated limb at the amputation site. These results suggested that PP2Aa played a crucial role in promoting limb regeneration by regulating cell proliferation. As a widely-distributed and evolutionarily conserved serine/threonine phosphatase, PP2A may serve as a key regulator in this process. These findings lay the groundwork for further exploration of the limited regenerative capacity of higher mammals.
