      爪哇虫草菌Cordyceps javanica是一种广谱性生防真菌,可防治多种农林害虫。效应因子在真菌致病寄主昆虫过程中发挥重要作用。本研究基于公布的爪哇虫草菌全基因组信息,对其中预测的11 142个蛋白序列进行筛选,以寻找其候选效应因子并进行功能分析。通过依次使用SignalP v5.0、TMHMM v2.0、TargetP v2.0和Protcomp v9.0等生物信息学工具进行预测,筛选出611个可能位于细胞质中的蛋白,并进一步分析了其中348个半胱氨酸含量较高的蛋白序列及117个具有串联重复单元的蛋白。通过比对病原物-寄主互作数据库(PHI-base),从鉴定出的57个具有注释的蛋白筛选出10个候选效应因子。最终,基于突变表型预测,筛选出4个与致病性相关的细胞质效应因子,并通过实时荧光定量PCR技术研究了两个效应因子编码基因(细胞壁蛋白TQW04886和疏水蛋白TQW02068)在爪哇虫草菌营养至生殖生长转变过程中的表达特征,结果表明这两个效应因子主要在产孢阶段高表达。本研究不仅为深入理解爪哇虫草菌与寄主昆虫互作的分子机制提供了基础,也为其他昆虫病原真菌效应因子的预测提供了参考。
英文摘要:
      Cordyceps javanica, a broad-spectrum insecticidal fungus, is extensively utilized to counteract agricultural and forest pests. Effectors are crucial in the interactions between entomopathogenic fungi and their host insects. Candidate effectors were predicted and analyzed from 11142 proteins in the whole genome of C. javanica using bioinformatics software, including SignalP v5.0, TMHMM v2.0, TargetP v2.0, and Protcomp v9.0. A total of 611 protein sequences were localized in the cytoplasm. Subsequent screening unveiled 348 sequences with a high cysteine content and 117 proteins featuring repeated sequences. A total of 57 protein sequences were compared with the pathogen-host interaction (PHI) database and identified. Protein sequences of less than 300 amino acids were selected, and among these, 10 candidate effectors were identified. Finally, four proteins were selected as putative effectors from C. javanica based on mutant phenotypes. The expression patterns of genes encoding secreted proteins (the cell wall protein TQW04886 and the hydrophobic proteinTQW02068) during the transition from vegetative to reproductive growth were investigated by quantitative real-time PCR. The results showed that these two genes were highly expressed in C. javanica during the sporulation stage. This study lays the foundation for a better understanding of the role of effectors in the interaction between the pathogen C. javanica and its host, and provides reference for the prediction and analysis of effectors in other entomopathogenic fungi.
