玉米象Sitophilus zeamais是一种世界性的储粮害虫,但其基因信息尚不完善。本研究利用高通量测序平台Illumina HiSeq TM2000对玉米象成虫进行转录组测序,总计获得64 358条unigenes,总长度39 481 752 bp,最短201 bp,最长29 046 bp,平均长度613 bp。将unigenes序列在7大数据库Nr、GO、Swissprot、KOG、Pfam、KEGG、TrEMBL中进行注释,分别有25 271、20 747、16 477、12 060、9 471、3 370、25 500条unigenes获得注释。通过GO功能分类,共有20 747条unigenes在GO数据库中生物学过程、细胞组分、分子功能3大类68个亚类功能组中找到对应。KOG结果显示,12 606条unigenes归到25个基因家族。通过KEGG代谢通路分析,共有3 370条 unigenes被注释,分别归属于细胞进程、环境信息进程、遗传信息进程、新陈代谢和有机体系统5大类代谢途径,共33个亚类274个功能通路。进一步的数据分析,共鉴定出146 081个SNP位点和5 002个简单序列重复(SSR)位点。本研究获得的玉米象转录组信息,为玉米象的功能基因挖掘提供了重要的信息资源。 |

Sitophilus zeamais is a worldwide grain storage pest, but its gene resources are deficient. The transcriptome of S. zeamais was sequenced using an Illumina HiSeq TM2000 platform. In total, 64 358 unigenes were obtained, with a total length of 39 481 752 bp, the shortest 201 bp, the longest 29 046 bp, and the average length of 613 bp. The unigenes sequence was annotated in 7 databases Nr, GO, Swissprot, KOG, Pfam, KEGG and TrEMBL, and 25 271, 20 747, 16 477, 12 060, 9 471, 3 370 and 25 500 unigenes were respectively annotated. By GO function annotation and classification, these unigenes can be divided into 68 subclasses in 3 categories: Biological process, cellular component and molecular function. Unigenes were further annotated based on KOG category,12 606 unigenes were annotated,which could be grouped into 25 functional categories. In KEGG pathways identification, 3 370 sequences were annotated, respectively belong to 5 kinds of cellular processes, enviromental in formation process,genetic information process,metabolic and organism system, which can be broadly divided into 33 subclasses, including 274 functional pathways. Further data analysis identified 146 081 SNP sites and 5 002 simple sequence repetition(SSR)sites. The transcriptome information obtained in this study provided important information resources for functional gene mining of S. zeamais. |
