| 【Aim】In recent years, the red imported fire ant, Solenopsis invicta, extensively invaded agricultural, forest, and urban habitats in China and posed serious threats to agricultural production, ecological security, and public health. During preliminary surveys of fire ants in nurseries in Guangdong Province, we learned from local forest farmers that certain herbicides might also exhibit insecticidal and repellent activities against S. invicta, thereby potentially providing both weed control and fire ant suppression.【Methods】In this study, three commonly used herbicides (triclopyr-2-butoxyethyl ester, picloram, and fomesafen) were selected to evaluate their effects on the survival and behavior of S. invicta, and transcriptomic analysis was employed to preliminarily explore the underlying molecular mechanisms.【Results】Triclopyr-2-butoxyethyl ester exhibited high toxicity to S. invicta workers, with median lethal dose (LD50) values of 2.407 μg/ant, 1.509 μg/ant and 1.411 μg/ant at 24 h, 48 h and 72 h, respectively. Additionally, at concentrations of 5 mg/kg, 50 mg/kg and 500 mg/kg, triclopyr-2-butoxyethyl ester significantly inhibited the digging behavior of S. invicta workers but had a relatively weak effect on foraging behavior. This was likely because foraging S. invicta workers transported large amounts of soil particles onto the herbicide-treated surface, thereby reducing their direct contact with the herbicide. In contrast, at the tested concentrations (5 mg/kg, 50 mg/kg and 500 mg/kg), picloram and fomesafen had no significant effects on the survival or behavior of S. invicta workers. Transcriptomic analysis revealed significant upregulation of some CYP450 genes and significant downregulation of olfactory-related genes and basal cellular function pathways in S. invicta workers following treatment with triclopyr-2-butoxyethyl ester.【Conclusion】Triclopyr-2-butoxyethyl ester exhibited marked toxicity and repellent activity against S. invicta workers, whereas picloram and fomesafen were likely to have relatively low non-target effects on ants. This study suggested that the application of triclopyr-2-butoxyethyl ester in agricultural and forestry habitats could simultaneously achieve weed control and fire ant management, thereby reducing management costs. |
