Research progress in the regulatory mechanism of insect oogenesis

  • 摘要:

    卵子发生是昆虫生殖过程中的关键环节,它不仅确保了种群的繁衍和遗传信息的传递,还与昆虫的适应性进化和社会结构的形成密切相关。本文概述了调控卵子发生的机制,包括形态学变化、功能意义以及多种影响因素。形态学上,卵子发生涉及从原始生殖细胞发育为成熟卵母细胞的连续过程,这一过程在不同昆虫中有所差异。影响因素上包括激素调控机制的多样性、环境和营养因素如何精细调控生殖过程,以及非激素因素如MicroRNA、转录因子、免疫信号和微生物代谢在卵子发生中的作用。内分泌激素,特别是保幼激素和蜕皮激素,在卵子发生中发挥核心作用,它们通过调节生殖干细胞的自我更新、卵母细胞成熟和营养供给等关键环节来调控卵子发生。不同昆虫中这些激素的作用机制存在差异,反映了昆虫在生殖调控上的多样性。除了激素调控,环境和营养因素也通过影响内分泌或神经系统对卵子发生产生影响。气候变化、营养状况等外部条件可以改变昆虫的生殖策略,从而影响种群的适应性和生存。此外,MicroRNA和转录因子在基因表达调控和细胞命运决定中扮演关键角色,而免疫信号和微生物代谢与卵子发生的相互作用也逐渐成为研究的焦点。本文旨在为理解昆虫卵子发生的复杂调控网络提供了全面的视角,并指出了未来研究潜在方向。

     

    Abstract:

    Oogenesis serves as a critical component in the reproductive cycle of insects, ensuring not only the perpetuation of species and the transmission of genetic information but also playing a significant role in the adaptive evolution and the establishment of social structures within insect populations. This article outlines the regulatory mechanisms of oogenesis, encompassing morphological transformations, functional implications, and a multitude of influencing factors. Morphologically, oogenesis entails a continuous developmental process from primordial germ cells to mature oocytes, which varies among different insect species. Influencing factors include the diversity of hormonal regulatory mechanisms, the nuanced control of reproductive processes by environmental and nutritional factors, and the roles of non-hormonal elements such as MicroRNAs, transcription factors, immune signals, and microbial metabolism in oogenesis. Endocrine hormones, particularly juvenile hormone and ecdysteroids, exert a central influence on oogenesis by modulating key stages such as germline stem cell self-renewal, oocyte maturation, and nutrient provision. The mechanisms of action of these hormones differ across species, reflecting the diversity in insect reproductive regulation. Beyond hormonal control, environmental and nutritional factors also impact oogenesis by affecting the endocrine or nervous systems. External conditions like climate change and nutritional status can alter reproductive strategies in insects, thereby influencing the adaptability and survival of populations. Additionally, MicroRNAs and transcription factors play pivotal roles in the regulation of gene expression and cell fate determination, while the interplay between immune signaling and microbial metabolism with oogenesis is gaining research interest. Consequently, this article aims to provide a comprehensive perspective for understanding the intricate regulatory network of insect oogenesis and points toward potential directions for future research.

     

  • 昆虫卵子发生(Oogenesis)的研究始于20世纪初,随着分子生物学和遗传学技术的发展,该领域取得了显著进展。早期研究主要集中在昆虫的解剖和组织学描述上,而近几十年的研究转向了分子机制和遗传调控(Hegner,1917;彭晨星等,2017)。具有里程碑性质的事件包括对特定昆虫(如黑腹果蝇Drosophila melanogaster)卵母细胞的形成与成熟过程的详细描述及其相关基因与信号通路的鉴定(奚耕思等,2010;Roy et al.,2017)。这些发现为人们提供解读生物多样性、昆虫行为及生态系统功能的新视角,并在疾病控制、生态保护及生物技术创新等多个领域开辟了应用前景。

    目前,卵子发生的研究借助新兴的技术手段(如基因编辑技术)为人们重新定义了生殖调控的分子机理(Chaverra-Rodriguez et al.,2018;Li et al.,2020),而环境因素对卵子发生的作用研究则进一步揭示了昆虫与其生活环境的复杂互动(Kambysellis et al.,1980;Wheeler,1996;Lenaerts et al.,2019)。同时,昆虫与微生物间的相互作用也正在揭示新的调控生殖的生物学机制(Otti,2015;Jordan and Tomberlin,2021)。这些领域的突破不仅推动了基础科学的深入,还引发了人们对昆虫种群管理的新策略的思考。

    卵子发生过程,涉及卵母细胞从形成到发育成为成熟的、可进行受精的卵的全过程。这一过程不仅是种群延续和基因传递的生物学基础,还是昆虫适应性进化、社会结构形成和环境感知的关键环节(Heming,2018;Church et al.,2021)。该过程涉及多样性细胞相互作用、精确性内分泌分子相互调节、时空特异性基因表达调控和严格控制性细胞周期运转等重要的生物学事件。因此,本文旨在总结当前有关卵子发生调控的研究进展,为未来相关领域的探索提供参考。

    卵子发生的形态学变化是一个复杂的过程,涉及到昆虫的生殖器官、卵子发生发育过程及产卵行为等。针对这些方面,前面已经有多名研究者对其进行了较为系统全面的阐述(Bonhag,1958;Engelmann,2013;Hoffmann,2018;Cury et al.,2019)。由于不同种类昆虫的卵子发生涉及的形态学变化不尽相同,本文以模式生物黑腹果蝇做个一般性的概括(图 1):首先,胚胎发育期的原始生殖细胞(Primordial germ cells,PGCs)和体细胞性腺前体细胞(Somatic gonadal precursors,SGPs)的产生与分裂形成卵巢结构。卵巢最前端被称为卵原区(Germarium),原卵区紧挨着体细胞性腺前体细胞分裂分化形成的端丝细胞(Terminal filament cells,TFs),而原卵区由包括帽细胞(Cap cells,CpCs)、生殖干细胞(Germline stem cells,GSCs)、内鞘细胞(Inner sheath cells,ISCs)、护送细胞(Escort cells,ECs)和包囊细胞(Cystoblast,CYB)等几类重要细胞组成。其中,包囊母细胞源于生殖干细胞的不对称分裂,包囊细胞再通过几轮胞质不对称分裂形成16细胞包囊细胞复合体(16-cell cyst),复合体细胞的其中1个细胞最终发育成为成熟卵母细胞(Oocyte),另外15个细胞作为滋养细胞(Nurse cells,NCs)为卵母细胞提供营养物质。随后,这16细胞包囊细胞复合体被体细胞滤泡干细胞(Somatic stem cells,SSCs)所增殖分化的滤泡细胞(Follicle cells,FCs)包裹形成卵室(Egg chamber),卵室在经历14个发育时期之后形成由卵壳包裹的成熟卵母细胞。这14个发育时期中又以滤泡细胞的增殖分化现象最为明显和复杂,包括滤泡细胞分化形成4种不同形态的滤泡细胞(Stretched、Centripetal、Posterior和Main body follicle cells)、边界细胞(Border cell,BC)和极细胞(Polar cell,PC)。这几种细胞作用于成熟卵的形态构建(背呼吸器、卵孔等结构)、卵的极性和胚胎形成等生物学过程,它们具体的形态描述和功能也已经被报道(Bastock and St Johnston,2008;Gilboa,2015;彭晨星等,2017)。总之,昆虫的卵子发生是一个复杂的细胞增殖分化事件,各类细胞相互配合并各自演化最终确保卵母细胞的形成。

    卵子发生的功能研究或者说卵子发生对于昆虫本身而言具备怎样的意义呢?不同的研究者有不同的见解,本文总结了以下几个方面:其一,生殖干细胞与其周围微环境(Niche)相互协调保证雌性昆虫能够持续产生足够数量的卵母细胞;其二,包囊细胞不对称胞质分裂并确定仅有1个细胞发育成为卵母细胞而其余15个细胞形成滋养细胞,从而保证了卵母细胞的质量;其三,卵母细胞通过减数分裂形成单倍体细胞,为后续的受精做准备,同时也保证了遗传物质传递的稳定性;其四,卵母细胞后期吸收卵黄蛋白等营养物质为胚胎发育提供物质基础;其五,卵壳等胞外结构的形成为卵子体外生存提供了保障(彭晨星等,2017)。因此可以大致地将卵子发生的功能意义分为以下三个部分进行研究。

    有学者将原卵区最前端这种由CpCs和前ISCs通过Dpp/BMP介导的信号传导和E-cadherin介导的细胞粘附形成控制生殖干细胞自我更新的区域称为生殖干细胞微环境(Microenvironment or niche)(Xie and Spradling,1998;Song et al.,2002;Song et al.,2004)。一般情况下,当GSC分裂时,一个由于保持与CpC的接触而维持自我更新,而新生成的子细胞则远离帽细胞并分化为CB。多个试验已经探究了这种由于空间特殊位置上的不同导致细胞命运不同的原因:第一,TF和CpC中的Piwi和Yb能够直接影响GSC自我更新的维持,同时Piwi能够影响干细胞的分裂速率(Cox et al.,1998;King and Lin,1999;Cox et al.,2000;King et al.,2001)。有趣的是,最近的研究指出piRNA和PIWI蛋白Aubergine(Aub)有助于激活果蝇生殖干细胞的糖酵解。糖酵解是GSC自我更新所必需的,在Aub功能丧失的情况下会诱导GSC的代谢转换,导致其分化(Rojas-Ríos et al.,2023);第二,CpC能够产生BMP信号,该信号能够直接通过生殖干细胞上的信号受体而控制其自我更新(Xie and Spradling,1998;Song et al.,2004),但BMP信号是一种短程信号,仅能过够在紧靠微环境的几个细胞中传递,当GSC的子代细胞远离该信号传递范围则无法维持自我更新的能力(Wilcockson et al.,2017);第三,CpC的数量与GSC的数量相关,当CpC包括直接接触GSC的CpC丢失后,GSC就会跟着缺失,而与TF却无关(Wang et al.,2011)。另外一项相近研究也指出由CpC和ISC等构成的微环境的大小会影响生殖干细胞的数量(Hsu et al.,2019)。这些证据表明,微环境不仅作为GSC的形成空间,也为维持其干细胞的特性提供了分子基础。除以上BMP信号通路和piRNA信号通路外,Notch信号通路、JAK/STAT信号通路、Wnt信号通路和Hedgehog(Hh)信号通路也被报道参与了生殖干细胞和微环境之间的协调作用(Forbes et al.,1996;Wang et al.,2008;Waghmare et al.,2018;Chen et al.,2023)。通过以上调控网络的相互配合确保产生了足够多且质量有保障的雌性昆虫生殖干细胞。

    生殖干细胞通过分化和分裂形成包囊细胞,包囊细胞形成的滋养细胞通过一种称为“环带孔”(Ring canals)的特殊细胞结构与卵母细胞连接。环带孔是在卵母细胞和滋养细胞的原始分裂过程中形成的胞质桥,这些胞质桥在细胞分裂时不会完全分离,从而保持了这些细胞之间的连通。环带孔的存在使得滋养细胞能够直接将胞浆、核糖体、mRNA、蛋白质及其它重要物质有效地输送到卵母细胞中。这种物质的直接转移对于卵母细胞的生长、发育即最终的成熟至关重要。通过这种方式,滋养细胞支持和促进了卵母细胞的生长,并确保了营养物质和遗传物质的适当供给。Notch、JAK/STAT、胰岛素及PKB/Akt信号通路等被发现参与调控了该过程(Irles et al.,2016;Das and Arur,2017;Borensztejn et al.,2018;Kalous et al.,2023)。

    果蝇卵母细胞减数分裂的启动通常需要卵母细胞达到一定的成熟度和大小之后才能发生,这一点通常在卵室的第3阶段开始,并持续到第14阶段。具体到减数分裂的精确启动,通常见于卵室的第6到第7阶段(Jia et al.,2015)。在这过程中通常需要卵母细胞在细胞质中建立极性,同时激活周期蛋白和周期依赖性蛋白激酶复合体来推动减数分裂的顺利进行(Bastock and St Johnston,2008)。这一过程还伴随着孢子形成检查点(Spindle assembly checkpoint,SAC)和DNA损伤修复机制的出现(Sun and kim,2012)。这些信号和监控机制共同确保了果蝇卵母细胞在减数分裂期间的正确分裂和发育,防止基因异常和染色体错误分配的风险,从而保障了后代的遗传稳定性。

    最后卵母细胞通过吸收卵黄原蛋白(Vitellogenin,Vg)和形成卵壳等胞外结构共同保证卵子后期的正常发育保障。昆虫卵子成熟过程包括卵母细胞对Vg的吸收,进而合成并储存卵黄蛋白以积累营养物质。因此,卵子的发生可分为3个阶段:卵黄形成前期(Previtellogenesis)、卵黄形成期(Vitellogenesis)和卵壳/绒毛膜形成期(Choriogenesis)。在昆虫体内,Vg作为主要的卵黄蛋白前体,在激素控制下主要在脂肪体中产生,并释放到血淋巴中。卵母细胞通过卵黄原蛋白受体(Vitellogenin receptor,VgR)和卵黄蛋白受体(Yolk protein receptor,YPR)介导的胞吞作用与内吞作用进行吸收。Vg及卵黄蛋白的合成与运输对卵巢的激活、卵黄生成及卵子的形成有着至关重要的影响。

    从前面卵子发生的形态学变化和功能意义上可以知道,卵子发生过程是一个高度复杂的过程,受到卵巢内部自身和外部因素的共同调控,这些因素包括内分泌激素、环境或营养信号、MicroRNA或转录因子及免疫信号与微生物代谢等(图 2)。

    内分泌激素(Endocrine hormones)是指由内分泌腺或内分泌细胞合成和分泌的化学物质,这些物质通过血液或其他体液传递到目标器官或组织,调节其生理功能。而参与昆虫卵子发生的内分泌激素主要有保幼激素(Juvenile hormone,JH)、蜕皮激素(Ecdysone)、神经激素(Neurohormones)、胰岛素样肽(Insulin-like peptides,ILPs)和促卵激素(Gonadotropic Hormones)等(Lenaerts et al.,2019;Manière et al.,2009;Santos et al.,2019;Swevers,2019)。其中,JH和蜕皮激素的活性物质20-羟基蜕皮激素(20-hydroxyecdysone,20E)是目前在昆虫卵子发生研究中最为热门的两种激素。JH和20E在控制卵子发生事件中的作用在不同种类昆虫中的作用是不同的。大体上,JH是不完全变态发育和部分完全变态发育的主要生殖调节激素,而20E则是部分膜翅目和鳞翅目以及全部双翅目的主要生殖调节激素(Roy et al.,2018;Santos et al.,2019)。由于技术手段不足等原因,早期对于JH和20E的研究主要集中于卵黄发生中后期即诱导或增强卵黄原蛋白表达及促进卵母细胞发育成熟与促进排卵行为等研究。具体研究显示,JH在黑腹果蝇、埃及伊蚊Aedes aegypti、赤拟谷盗Tribolium castaneum和飞蝗Locusta migratoria等昆虫中通过多个途径促进Vg的合成和卵母细胞对Vg的摄取,以此促进卵母细胞发育(Sheng et al.,2011;Clifton and Noriega,2012;Song et al.,2014)。对于JH的作用靶点与下游基因,学者们普遍支持MEKRE93途径,即JH与受体Methoprene-tolerant(Met)蛋白结合并级联下游反应,该级联的下游成员包括Krüppel同源物1(Kr-h1)和E93等(Belles and Santos,2014)。值得注意的是,当果蝇卵巢微环境中的帽细胞JH信号被阻断之后,会导致生殖干细胞数量的减少(Luo et al.,2020),表明JH信号并不像过去所报道地仅仅影响卵子发生的中后期,还可能存在未知的机制来影响卵子发生前期各类细胞的增殖与分化。

    同样的,在黑腹果蝇、埃及伊蚊和赤拟谷盗的研究中也发现20E参与了协调Vg相关调控基因的表达和卵母细胞后期的发育(Roy et al.,2018)。在棉铃虫Helicoverpa armigera、家蚕Bombyx mori和斜纹夜蛾Spodoptera litura等昆虫研究中发现20E还参与了卵巢组织细胞的自噬和凋亡,表明20E在卵子发生过程中具有重要作用(Lin et al.,2023)。此外,20E也被证明在卵黄发生前期即卵室形成之前发挥重要作用。首先,在幼虫发育前期,20E能够抑制果蝇卵巢微环境和原始生殖干细胞的分化,而在幼虫发育后期则是促进生殖干细胞的增殖,并在生殖干细胞自我更新的维持上发挥重要作用,同时也是包囊细胞复合体发育和滤泡细胞发育所必须的(Belles and Piulachs,2015)。而相关的具体调节机制在Swevers等人所报道的综述中有所体现(Swevers,2019;Rumbo et al.,2023)。因此JH和20E都可能在卵子发生地整个过程中发挥重要作用,但不同昆虫物种侧重点有所不同,后续研究中或许能够从激素脉冲信号和各类激素调控节点入手,进一步加深相关研究。

    还有一些有趣的报道称,大猿叶虫Colaphellus bowringi个体感知光周期信号可以改变内分泌信号如蜕皮激素信号等,以指导繁殖滞育特征,如卵巢停滞和脂肪积累增加等(Guo et al.,2021)。无独有偶,在对家蚕响应昼夜变化时的研究表明,蜕皮激素和保幼激素的变化也能使5龄期幼虫发育减慢(Qiu et al.,2023)。这些报道虽未具体阐明内分泌信号是如何响应外界环境信号的具体分子机制,但此类发现无疑增加了人们探索昆虫繁殖调控途径的兴趣,更是扩大了人们对于内分泌激素在昆虫卵子发生中作用的认知。

    上述提到大猿叶虫感知光周期可以通过内分泌信号进而影响卵子发生的进程,那么是否还有其它环境能够影响昆虫的卵子发生呢?我们知道昆虫的迁飞行为是一种对昆虫种群生存和繁殖的一种重大策略,而迁飞的目的就是为了获得更加合适的环境与后代生长发育所需的营养,因此,气候变化和季节变化便通过迁飞行为对昆虫卵子发生产生影响。研究表明,一方面迁飞行为会造成昆虫产卵量少、卵巢发育慢及交配延迟,另一方面迁飞行为又能刺激卵母细胞生长和产卵的提前(Highnam and Haskell,1964;Rankin et al.,1986;Kim and You,2022)。在这一过程中,环境中的营养因素起到最为重要的作用。在昆虫卵子发生期间,胰岛素样生长因子(Insulin-like peptides,ILPs)参与调节营养物质的分配,胰岛素样生长因子能够感应营养状况并调节相应的生理反应,调整营养物质向卵巢的分配,从而影响卵子的质量和数量,若昆虫所获得营养不足,会导致滞育现象发生(Drummond-Barbosa and Spradling,2001;Sim and Denlinger,2013)。同时,在社会分工较为明确的种群中(如蚂蚁),胰岛素样生长因子还与不同种类蚂蚁的生殖分化相关,即蚁后具备生育能力而工蚁在发育后期丧失生育能力(Okada et al.,2010)。在最新的研究中发现,柑橘木虱Diaphorina citri取食不同(嫩叶和老叶)同样可能通过胰岛素样生长因子通路造成雌性卵巢发育不同,进而影响产卵(Wang et al.,2023)。因此,营养是否充足、营养来源及营养种类在昆虫卵子发生中具有显著的作用。当然,胰岛素样生长因子并不是作为平衡机体和性腺营养分配的唯一途径,例如在对黑腹果蝇唾液腺的miR-1000基因的研究发现,沉默该基因能够影响雌性个体的进食效率,从而影响卵巢的发育(Rahman et al.,2020),表明昆虫取食与否和后面对营养的分配受到胰岛素样生长因子和其他机制的共同调控。而在我们实际生产生活中,家蚕在低温饲养条件下往往也出现滞育卵,说明温度变化也同样影响着昆虫的繁殖。因此,环境因素和营养条件共同为昆虫卵子发生和繁殖提供了外在条件,而探索这些外在因素与内在分子之间的关键节点或者称为靶标分子尤为关键。

    MicroRNA(miRNA)是一类内生的、长度约20~24个核苷酸的小RNA分子,与靶mRNA的3'-非翻译区或编码序列互补结合,以调节转录后水平的基因表达(Forman et al.,2008;Rigoutsos,2009;Ghildiyal et al.,2010),而转录因子则是一类能够与DNA特定区域结合的蛋白,在基因表达调控中起重要作用。因此,在对昆虫卵子发生过程中的MicroRNA或转录因子的研究也越来越多。研究发现,埃及伊蚊的miR-275和miR-305受到抑制后会导致卵子发育不完全,miR-277和miR-309的敲除也能使其卵泡发育异常卵巢生长缺陷,而蚊子特异的miR-1890也能通过靶向丝氨酸蛋白酶JHA15影响卵巢的发育(Bryant et al.,2010;Lucas et al.,2015;Zhang et al.,2016;Ling et al.,2017)。在棉铃虫中的研究也发现miR-2002b的喂食也降低了成虫的生殖力(Ramu et al.,2018)。而对于MicroRNA介导昆虫卵子发生的具体机制研究中发现,蝗虫的miR-278可以靶向早期的JH应答基因从而导致Vg降解并进一步阻断卵母细胞成熟和卵巢生长(Song et al.,2018)。另外一项重要的研究报道发现,敲除Dcr-1(一种miRNA加工酶)后果蝇卵巢中的生殖干细胞无法维持并迅速消失,表明MicroRNA在生殖干细胞维持中的重要作用(Jin and Xie,2007)。

    而相关转录因子的报道较多,例如上文提到的JH与受体Methoprene-tolerant(Met)蛋白参与的MEKRE93级联调控途径中,Kr-h1和E93等均可作为转录因子发挥作用。另外埃及伊蚊血食后20E信号刺激早期效应因子如BrC-Z2、βFTZ-F1、E74B和E75A的表达,而后这些效应因子作为Vg合成相关基因的转录因子发挥作用,从而促进卵子发生(Ros et al.,2018)。同时值得注意的是,这些转录因子不仅仅参与转录激活,同时参与了卵子发生相关基因的转录抑制,多篇综述文章概括了转录因子及转录因子与MicroRNA的相互作用在昆虫卵子发生中的重要作用,在这里便不过多介绍(Lenaerts et al.,2019;Santos et al.,2019;Swevers,2019;Song and Zhou,2020;Wu et al.,2021)。

    近年来,免疫信号和微生物代谢对于昆虫卵子的发生的影响成为了研究热点。在过去,免疫应答作为机体抵御病原体和寄生虫的入侵的功能所熟知,但研究者发现免疫激活状态下,昆虫体内的资源分配会发生改变,以优先支持免疫防御机制,这可能会导致卵巢发育受到影响。研究显示,果蝇在受到细菌感染后,会通过Toll和IMD途径激活免疫反应,这会导致卵巢发育的延迟或抑制(Schwenke et al.,2016)。感染疟原虫的蚊子是卵巢发育会受到影响,卵子产量减少(Hurd,2003)。在豌豆蚜Acyrthosiphon pisum的研究中也显示,其共生菌Regiella insecticola能够提高产卵量(Tsuchida et al.,2011)。这些研究表明,免疫信号与昆虫的卵子发生之间存在着复杂的相互作用,免疫应答的激活可能会通过不同的机制影响昆虫的生殖系统和繁殖策略。然而,免疫信号对卵子发生的具体影响可能因昆虫种类、感染类型及环境条件的不同而有所差异(Nunes et al.,2021)。

    同时微生物也能够以多种方式影响昆虫的卵子发生。这些影响可能是直接的,例如通过感染昆虫的生殖系统的试验证明果蝇在遭受细菌感染后,会出现卵巢大小减小和卵子产量下降的现象(Schwenke et al.,2016)。这些影响也可能是间接的,例如通过影响昆虫的整体健康状况、营养状况或免疫系统来间接影响卵子的发育和成熟。研究发现,沃尔巴克菌Wolbachia能够通过影响昆虫的生殖系统来促进自身的传播,包括引起雌性生殖器官中的卵子发育异常(Werren et al.,2008)。另外在某些植食性昆虫中,共生细菌的存在对于宿主合成必需氨基酸是必不可少的,这直接关系到昆虫的生殖成功率(Douglas,2009)。这些研究虽未阐明昆虫在做出免疫应答及面对微生物入侵时如何分配体内资源并调控卵子发生的机制,但也从正面回答了这些因素与卵子发生的相关性。病原和机体在长期相互演化中往往进化出了特定的机制,不同昆虫种群中寄居着不同的微生物类型,甚至在不同地区也有独特的依存和竞争关系,因此,从微生物群落分布特征和代谢物与昆虫自身表型变化相适应的角度入手,或许会成为该领域研究的方向。

    本文综述了昆虫卵子发生的调控机制,包括形态学变化、功能意义及影响其发生的多种因素。卵子发生的形态学变化在不同昆虫中有所差异,但通常涉及从原始生殖细胞到成熟卵母细胞的一系列复杂过程。卵子发生对昆虫而言至关重要,它不仅保证了种群的延续和基因的传递,还涉及昆虫的适应性进化和社会结构的形成。内分泌激素,尤其是JH和20E,在卵子发生中扮演着核心角色,它们通过影响生殖干细胞的自我更新、卵母细胞的成熟及营养物质的供给等方面发挥作用。但是JH和20E在不同昆虫中的作用是具有差异的,表明昆虫可能通过不同的激素调节机制来适应其特定的生殖需求且这种多样性可能与昆虫的卵母细胞发育过程、生活史和环境适应性有关。在这里需要注意的是,包括JH和20E在内的内分泌激素在调控卵子发生的过程中并不是单一激素发挥作用的,它们往往以相互协同或相互抑制的方式确保昆虫卵子发生全过程的顺利进行。研究表明在埃及伊蚊中,20E可以通过激活关键的JH合成酶进而影响卵子发生和卵巢发育(Ahmed et al.,2020)。在家蚕中的研究也表明,作为JH关键转录因子的Krüppel homolog 1(Kr-h1)也可以被20E信号通路上的转录因子BmKRP诱导表达(Zhu et al.,2021)。最近一篇关于内分泌激素在昆虫卵子发生中相互作用的综述也进行了多方面的描述(Leyria,2024)。总之内分泌激素之间的相互串扰共同制约和平衡着卵子发生的全过程,随着不同类型的内分泌激素的发现,尤其是神经肽类激素(Neuropeptides)的发现进一步拓展和增加了内分泌激素对卵子发生过程的调控网络。同时环境和营养因素,如光周期和营养状况,也通过内分泌信号或神经信号对卵子发生产生影响,且在如今全球气候变化和环境生态改变的背景下,了解环境和营养因素对昆虫生殖调控的机制将成为预测昆虫种群动态和制定有效害虫管理策略的关键一环。此外,MicroRNA和转录因子在调控基因表达和细胞命运决定中起着重要作用,这些分子的调控网络可能非常复杂,涉及多个层面的相互作用。尤其是MicroRNA、转录因子、内分泌激素、环境营养信号等多因素的相互作用对卵子发生过程的精确调控。现如今已经有相关报道表明橘小实蝇Bactrocera dorsalis的MicroRNA、转录因子和JH信号之间的关系,即受JH控制的miR-309a可以结合转录因子Pannier(GATA转录因子A/pnr),激活卵黄蛋白2和卵黄蛋白受体的表达,从而促进卵巢发育(Zhang et al.,2022)。未来的研究可以利用高通量测序技术和计算生物学方法来揭示这些调控网络的全貌,并探索它们在生殖发育异常中的作用。最后,免疫信号和微生物代谢对卵子发生的影响也日益受到关注,它们通过改变昆虫体内的资源分配来影响生殖系统和繁殖策略,这种权衡可能涉及到复杂的生理和行为适应。深入研究这种权衡的分子机制,不仅有助于我们理解昆虫的生态和进化策略,也可能为疾病控制和生物保护提供新的视角,同时探索微生物如何通过代谢途径影响宿主的生殖过程,也有助于开发新的生物学技术用于害虫防治或生物修复。尽管我们已经开展了大量卵子发生调控机制的相关研究,但目前有效应用于现实生活中防控害虫和利用昆虫生产的技术手段依旧有限,因此我们应该采取多学科交叉融合的方式,借助化学手段和物理手段以及新兴的人工智能手段,在更高的维度上统一(生态环境—昆虫种群—昆虫个体—卵巢组织—生殖细胞)这一层级链,在已有的研究基础上,清晰且明确的规化出一条或者几条研究发展路线,并在实际生产生活中实践和总结,以此达到加速该领域研究和发展的目的。
