Hailed as the“Third Agricultural Pesticide Revolution”, RNA pesticides, a category of nucleic acid pesticides based on RNA interference (RNAi), hold broad application prospects in the field of pest control. At present, relevant research on pest management using RNA pesticides mainly focuses on several technical approaches, including gene silencing mediated by chemically synthesized double-stranded RNA (dsRNA) or microRNA (miRNA) in vitro, host-induced gene silencing (HIGS) mediated by transgenic crops, microbe-induced gene silencing (MIGS), and virus-induced gene silencing (VIGS). In the insect RNAi-based pest control system, the optimization of delivery systems and improvement of stability for dsRNA and miRNA remain core bottlenecks to be addressed. To tackle this technical challenge, the strategy of constructing composite delivery systems via the co-assembly of nanomaterials as carriers with dsRNA and miRNA has attracted extensive attention from both academic and industrial communities in recent years. This nanocomposite delivery system exhibits both biosecurity and high delivery efficiency: it can not only effectively protect nucleic acid molecules from degradation by environmental nucleases, but also significantly enhance the RNAi knockdown efficiency of dsRNA and miRNA, thus showing great application potential in the field of green pest control. This paper systematically summarizes the research progress and application status of RNA interference technology in pest control, aiming to provide a reference for the applied research and industrialization of RNA pesticides.

RNA interference (RNAi) is a gene silencing phenomenon caused by double-stranded RNA (dsRNA), which has brought about significant changes to the field of biology, and demonstrated great value in various areas such as gene function research, medicine, and agriculture. The efficiency of RNAi in interfering with target genes is influenced by multiple factors, among which the delivery method of dsRNA plays a crucial role. In entomological research, dsRNA can be delivered through methods such as injection, feeding, cuticle penetration (soaking or spraying), and transgenic plant-mediated delivery, enabling the identification of gene functions or other application developments. This article reviews these delivery methods and analyzes some key factors affecting dsRNA delivery efficiency, in order to provide references for numerous entomological studies and the application methodologies of this technology.

http://hjkcxb.alljournals.net/hjkcxben/ch/reader/view_abstract.aspx?file_no=202602004&flag=1

【Aim】The fall armyworm, Spodoptera frugiperda, is a major agricultural pest with high reproductive potential and poses a significant threat to a wide range of crops. Currently, it has developed resistance to multiple insecticides. Therefore, alternative green control strategies are urgently needed.【Methods】Previous research revealed that overexpression or inhibition of microRNA, miR-34-5p, resulted in high mortality in multiple lepidopteran pests such as Spodoptera frugiperda. To further evaluate the potential application of miR-34-5p in pest control, we constructed genetically modified maize expressing miR-34-5p. After the fall armyworm fed on this maize for 96 hours, the expression level of miR-34-5p significantly increased by 2.5 fold, and the expression level of its target gene, the ecdysteroid receptor SfEcR, significantly decreased by 45.5%. 【Results】Bioassay results demonstrated that the cumulative mortality of S. frugiperda fed on miR-34-5p transgenic maize were significantly increased by 25.0%~40.0%, compared with that of larvae fed on wild-type maize. Surviving larvae exhibited significant developmental defects and low fecundity.【Conclusion】These results indicate that insect microRNA-based transgenic crops exhibit promising control efficacy against S. frugiperda, which provides a theoretical basis and technical support for the development of miRNA-based green pest control strategies.

http://hjkcxb.alljournals.net/hjkcxben/ch/reader/view_abstract.aspx?file_no=202602005&flag=1

【Aim】SMARCE1, an essential component that forms the core of chromatin remodeling complexes, plays a significant role in various biological processes including gene expression regulation and cell division. To elucidate the functional role of SMARCE1 in the developmental process of Spodoptera litura, we cloned and characterized the SMARCE1 gene in this study. The protein it encodes consists of 570 amino acid protein residues.【Methods】Multiple sequence alignment and the phylogenetic analysis revealed that SlSMARCE1 protein exhibited a high degree of conservation and significant homology with SMARCE1 proteins in other insects. During S. litura larval development, SlSMARCE1 exhibited stage-specific expression with peak levels observed in third-instar larvae. Tissue-specific profiling in fifth-instar larvae revealed predominant SlSMARCE1 expression in the midgut. To verify the function of SlSMARCE1, this study investigated the characteristics of the gene in S. litura and its biological effects under RNA interference.【Results】The results showed that silencing of SlSMARCE1 by RNA interference significantly inhibited the growth and development, decreased larval weight and size of S. litura larvae during larval development. Furthermore, after spraying dsSlSMARCE1 to larvae at the end of the 1st instar, the survival rate of larvae was 55% within at 72 h, and the expression of Ago and Dicer, key genes of the RNAi pathway elevated significantly.【Conclusion】These results not only demonstrate that SlSMARCE1 has an important role in the growth and development of S. litura, but also provide new target genes for the prevention and control of the S. litura using RNAi technology, and provide a promising strategy for reducing the reliance on chemical insecticides in agricultural pest control.

【Aim】Calcineurin (CaN), a Ca2?-dependent serine/threonine protein phosphatase belonging to the protein phosphatase 2B (PP2B) family, plays diverse regulatory roles in various physiological and biochemical pathways in vivo.【Methods】To investigate its potential as a key target gene for RNA interference (RNAi)-based pesticide development, this study conducted CaN RNAi on 4th instar Helicoverpa armigera and statistically analyzed their mortality, body weight, pupation rate, eclosion rate and other life indicators.【Results】The results showed that injection of CaN dsRNA significantly reduced both CaN gene expression and the enzymatic activity of CaN protein. Following knocking down CaN expression, about 60.81% mortality was observed among the fourth-instar larvae. Moreover, the body weight of surviving larvae showed a significant reduction starting 24 h after CaN dsRNA administration. The developmental durations from 4th instar larva to pupa and from pupa to adult eclosion were significantly prolonged. Both pupation and eclosion rates were markedly decreased, with the final eclosion rate dropping to only 3.75%.【Conclusion】These findings demonstrate that CaN is essential for the growth and development of H. armigera and highlight its potential as a promising RNAi target for pest control strategies.
