Artificial identification and classification of Hyphantria Cunea in insect monitoring are time-consuming and laborious, and subjective. In this paper, the RPN artificial neural network model is used to extract the features of the Hyphantria Cunea image data, and the ResNet50 and ResNet101 network models are compared and analyzed. An improved artificial neural network Recognition model IHCDM of Hyphantria Cunea is designed (Improved Hyphantria Cunea Artificial Neural Network Recognition Model，IHCDM). The model is trained on GPU processor by end-to-end method, and verified by experiments. The results showed that the recognition accuracy of this model was 99.5%, which was 0.5% and 0.4% higher than that of ResNet50 and ResNet101. After Hyper-parameter adjustment, as if the confidence threshold was 0.75, the recognition accuracy was 99.7%, and the recognition speed was 0.09 ms/piece. The improved image recognition model provides a new method for the rapid identification and classification of Hyphantria Cunea.

We compared and analyzed the ultrastructure, type, distribution and number of antennal sensilla in morphs of the aphid Schlechtendalia chinensis (Bell), using scanning electron microscopy. The results showed that S.chinensis has four types of antennal sensilla: Trichoid sensilla, primary rhinaria, sensory projections and secondary rhinaria, all of which varied in their distribution and number on the antenna in the different morphs. Trichoid sensilla could be divided into I and II types, and they distributed on all morphs of S.chinensis’antenna. Primary rhinaria were mainly observed on the antenna of fundatrices, apterous fundatrigeniae, nymphal alate fundatrigeniae, males and females. There are two types of PrⅠ and PrⅡ, occurring on the last and penultimate segment, respectively. But there was only PrⅠon the antenna of nymphal alate fundatrigeniae. The sensory projection was only located on the third to fifth segments of the antenna of nymphal alate fundatrigeniae. Secondary rhinaria were only located on the third to fifth segments of the antenna of alate fundatrigeniae and alate sexuparae. And they accounted for approximately two-thirds of the antennal surface. We suggest that these differences are related to the habitat of the particular morph (e.g. internal or external to the gall), its main form of behavior (e.g. feeding, mate selection) and whether the morph is winged (e.g. host or migration).

This study describes a method for accurately and quickly identifying the sex of the Cricula variabilis old-instar larvae, pupae and adults. And it compares the characteristics of adults and pupae, male and female. The female genital pore is located in the 8th and 9th abdominal segments, and the 8th abdominal segment is not closed. The male genital pore is located only on the 9th abdominal segment, and there are lumps on both sides of the genital pore. There are more light-colored areas near the ecdysial line of the female larvae's head shell, and there are fewer male light-colored areas. And the color of the female postclypeus area is also lighter than that of the male. The male and female adults differ greatly. The antennae of the male are pinnately shaped. The forewings are generally without transparent spots, or spots are small. The antennae of the female are short and pinnate which looks like filament, and the forewings have three large and continuous transparent spots. After measuring 30 pairsof pupae, it was found that there were significant differences between the male and female pupae (P＜0.05) in the morphological characteristics data such as pupae length, pupae width and pupae weight.

Megastigmus sabinae Xu et He is an oligotrophic pest that harms the seed kernels of Sabina przewalskii Kom. The larvae eat endosperm in the cones and develop synchronously with the cones, resulting in fruitless seeds after the seeds mature. Seriously affect the seed production of Sabina przewalskii Kom. To observe and analyze the morphological characteristics of the antennae, maxipalp and ovipositor receptors of the female adult of M. sabinae, aiming to provide a basis for the research on the evolution and behavior strategies of the larvae of M. sabinae. Scanning electron microscopy was used to observe the antennae, maxipalp and the receptors on the ovipositor of the female adult of M. sabinae. The antennae of M. sabinae are knee-shaped, consisting of 14 subsections, radicula, scape, pedicel, annellus, funicle(7 sections), clava(3 sections）. Total length 24~29 μm, with Bohm bristles, placoid sensilla, trichoid sensilla, chaetica sensilla, basiconic mastoid sensilla on the surface; the maxipalp composed of 5 sections, the first section is thick and long, and the remaining 4 sections is thin, it has four types of sensors: trichoid sensilla, chaetica sensilla, styloconic sensilla and basiconic sensilla; The external ovipositor is composed of ovipositor sheath and speculaspeculae. It has a trichoid sensilla, coeloconic sensilla, misphaericis sensilla and styloconic sensilla. The ovipositor sheath is 3.17±0.22 mm long, with 2 types of trichoid sensilla on the surface, and the specula speculae is 5.01±0.41 mm long. The specula speculae starts at the base and is divided into 3 valves, and the outer end has an “eight” shape Notch and 2 styloconic sensilla. The female receptors on antennae, maxipalp and ovipositors of M. sabinae are diversal and rich in morphological changes. There are certain differences between different types of receptors, which are the adaptive morphology produced by the small mole of M. sabinae to adapt to survival. This article is the first report on the antennae, maxipalp and ovipositor receptors, and provides a theoretical basis for understanding the morphological characteristics, evolution, and behavioral strategies of the small mollusk.

Huanglongbing is the most devastative disease in citrus production, early detection and diagnosis in a timely and rapid manner is one of the key measures to prevent and control huanglongbing. In this paper, the DNA sample carrying Candidatus Liberibacter asiaticus (CLas) was sequenced by MinION, and the sequencing results were compared to the genome of CLas by blast, graphmap, minimap2 and two BWA comparison methods. The comparison results were evenly compared to the genome of CLas, and no serious bias was found, which verified the reliability of the sequencing results. This sequencing technology could make up for the difficulty of optical identification due to the small or damaged Diaphorina citri (Kuwayama), and it could also detect whether the psyllids carry CLas at the same time, and provided real-time monitoring and early warning for citrus planting areas where there was a potential risk of huanglongbing.

In order to determine the rapid, nondestructive and accurate identification of male and female pupae of Apocheima cinerarius Erschoff pupae, external distinguishing features of head, thorax, abdomen and body colour of A. cinerarius pupae were analyzed, and the results of identification were verified by dissecting the reproductive system. The results showed that the accurate rate of distinguishing features of thorax and abdomen were obviously higher than that of head and body colour, which could reach 100%. Firstly, there was a Y-type sulcus in the middle of the 8th abdominal segment of female pupae, and the Y-type sulcus and the 7th abdominal segment trailing edge were combined to form an inverted triangle, but male pupae wasn't this characteristic. Secondly, there was a longitudinal suture gonopore in the middle of the 9th abdominal segment of male pupae, and the gonopore had a semicircular verrucae on each side, but the gonopore was connected with the oviposition aperture to form a suture with flat on both sides. Lastly, the relative length of each segment of thorax with tergum of male pupae was wider than that of female pupae, especially mesothorax with tergum of male pupae was the widest, and that of the trailing edge protrudes sharply outward. Therefore, the characteristics of thorax and abdomen of A. cinerarius pupae can quickly and accurately identified male and female pupae.

In order to explore the difference in collection efficiency of different insect capture methods, insects were collected using four sampling methods: The Malaise trap, the pitfall trap (mixture of sugar and acetic acid and water in the trap), the line transect and the light trap, in different habitat types in Zhenzhuquan Town, Yanqing District, Beijing. Insects collected using different collection methods in different habitat types were be analyzed. The results showed that 3 996 insects were collected in the experiments, belonging to 12 orders and 87 families. Among them, Lepidoptera, Hemiptera and Coleoptera were the most diverse species in insecta, and the number of Hemiptera, Diptera and Lepidoptera were the highest. The line transect collected 72 species of 61 families of insects, 95 species of 45 families by light trap, 41 species of 39 families collected by Malaise trap, and 25 species of 14 families by pitfall trap. In the experiment, there were significant differences in the number of insects collected by different methods in different habitats (P<0.05). The similarity coefficient between different collection methods except the Malaise trap and the line transect was low (q=0.436), and the other methods were lower. It is currently believed that the Malaise trap was the most preferred method of collection in survey, because the Malaise trap was the most convenient to collect insects and saves manpower; the line transect had the most stable effect; the light trapping method had good collection effect, but the application was more complicated. Compared with the other three methods, the efficiency of pitfall trap could not collect large number of insects or collect specific insects. If no special demand, it could be considered to reduce or abandon to use in survey.

It is one of the rare silk production insects that can be largely reared indoors, which can be reared in successive generations throughout the year. The large-scale indoor rearing production of Samia ricini may be ruined by destructive diseases which includes microspore disease, nuclear polyhedrosis disease, and soft rot disease. Diseases will become increasing serious with the continuous accumulation of pathogens, and the pathogens become more difficult to remove. Therefore, disease monitoring is the most important means to prevent the disease outbreak. MinION is a portable high-throughput sequencing platform, which is convenient for pathogen DNA sequencing. In this paper, the DNA of sick Samia ricini was sequenced by MinION, and the unknown pathogen DNA sequences were attempted to be identified. The feasibility of using MinION to identify the pathogen was discussed, which provided a guarantee for the monitoring of silkworm disease.

Antennae and labial palp are important sensory organ on the head of insects, usually covered with various types of sensilla. Protoschinia scutosa is an important agricultural pest, but the antennal and labial palp sensilla remain unsatisfactorily explored. This study investigated the types, distribution and ultramorphology of the antennal and labial palp sensilla of P. scutosa using scanning electron microscopy. The antennae of both sexes of P. scutosa were filiform, but female flagellum was significantly shorter than male. Both sexes bear eight types of sensilla on antennae: Bohm’s bristles, sensilla squamiformia, s. trichodea, s. chaetica, s. auricillica, s. basiconica, s. coeloconica, and s. styloconica. Sensilla trichodea were the most abundant type among these sensillum types, and males possessed much more sensilla trichodea than females. The labial palp of P. scutosa was long tubular, and showed non-significant differences in length between sexes. Five types of sensilla were observed on the labial palp, sensilla trichodea, s. campaniformia, s. squamiformia, s. chaetica and club-shaped sensilla. Sensilla trichodea were located on the basal segment of labial palp and the distal labial pit organ, s. campaniformia were distributed among the cluster of s. trichodea on the basal segment, s. squamiformia were present on all three segments, and s. chaetica and club-shaped sensilla were only found on the distal segment. This study briefly predicted and discussed the function of antennal and labial palp sensilla, and would provide a foundation for functional analysis of insect sensilla, taxonomy and behavioral studies.

Tuta absoluta is an oligophagous biology invasive pest, which extremely harm to tomato fields, and seriously threaten the tomato industry in Xinjiang, China. Because of the lack of information on the biology of T. absoluta and difficulty to feed in laboratory, our study aimed to determine the number of larval instars, and evaluate the effect of temperature on its development, survival and reproduction, and also optimize the artificial diet. The results showed that the larvae were divided into four instars according to both the head shell and the times of moulting. The results of the temperature treatments (20℃, 25℃, 30℃ and 35℃) showed that the whole larval stage of T. absoluta was 11.04 d at 25℃, which was shorter than that at 20℃, and longer than that at 30℃, and the larvae could not develop at 35℃. Both the survival rate and fecundity of T. absoluta at 25℃ were the highest in the different temperature treatment (20℃, 25℃, and 30℃), which indicated that 25℃ was the optimum temperature for its development. Besides, we designed three artificial diets to feed T. absoluta, the average number of eggs laid per female of T. absoluta feeding C diet was about 39, which was more than that of the two other, and its developmental duration (40 d) was shortest, which clarified that the C diet was a suitable artificial diet, which laid the foundation for indoor breeding of T. absoluta.

To better understand the mechanism of action of destruxin A (DA) on Bemisia tabaci (Gennadius) (Hemiptera∶Aleyrodidae), transmission electron microscopy (TEM) was used to observe the pathological changes in the midgut and fat body tissues of adult whiteflies fed with DA. The results showed that the midgut cells of the whiteflies were severely damaged after feeding with DA. The vacuolization of the cells accrued and the nuclear membrane were swelling in the early treatment stage. At the later stage of treatment, all the microvilli fell off and the contents of the organelles died out. The fat body cells of B. tabaci adults became lighter in color in the early stage of destruxin A treatment and the lipid droplet membrane structure became transparent. Moreover, a large number of vacuoles appeared. At the later stage of treatment, the lipid droplet membrane structure collapsed. The breakdown of midgut and fat body cells may be one of the important reasons for the death of whiteflies caused by DA. Our results providing a scientific basis for further research into the mechanism of DA.

This study aims to clarify the number of larval instars and to understand the types, morphology and distribution of sensilla on the instar larval head of Illidcris pruni Dyar. The body length and head capsule width of I.pruni larvae were measured. Based on the frequency histograms and Dyar's rule of the obtained data，the larval instars were determined. The obtained result was verified by Crosby growth rule and linear regression method. The morphology and sensilla on the head of the final instar larva of I.pruni were observed under scanning electron microscope. The results showed that the frequency histograms of the body length and head capsule width of I.pruni larvae showed 5 obvious peaks, and it is so inferred that I.pruni larvae have 5 instars. The 5 instars had the body length range of 2.96~4.04 mm, 5.27~6.27 mm, 7.76~8.36 mm, 9.47~10.45 mm and 11.83~12.11 mm, the head capsule width range of 0.316~0.358 mm, 0.397~0.459 mm, 0.538~0.598 mm, 0.687~0.735 mm and 0.947~1.011 mm. The head capsule width in accordance with Dyar's rule and Crosby growth rule, and showed obvious linear relationship. Therefore, the head capsule width could be used as important indexes for the division, body length could be used as an auxiliary index for the division of larval instars of I.pruni. Three types of sensilla including sensillum styloconicum, sensillum basiconicum and sensillum chaeticum were observed on the head and are mainly located on the antennae, mouthparts, mandible and maxillary palpus.

This paper armed to study the indoor rearing method of Chilo infuscatellus Snellen, which provided well-developed test insects for pesticide screening and pesticide toxicology, and to carry out deep research on the biology and ecology of C.infuscatellus was successive reared by corn cob in laboratory. The main life parameters were observed in the 1st, 4th, 7th and 10th generations, and the feeding effect was evaluated. The differences of development indexes and life parameters were observed in the 5th generation, sugarcane seedling+sugarcane stem section and corn cob+sugarcane stem section. The results showed that hatchability and eclosion rates of the 1st, 4th, 7th and 10th generations of C.infuscatellus fed with corn cob were all above 80%. The survival rates of larvae were between 40% to 60%, and the fecundity of single female was between 25 to 50 eggs. The hatching rate, larval survival rate and single female fecundity of corn cob feeding were higher than that of sugarcane seedling+sugarcane stem section and corn cob+sugarcane stem section feeding. The net reproductive capacity R0(26.17), the intrinsic growth rate rm (0.08) and the limited growth rate λ (1.09) of the population fed with corn cob were all higher than those of sugarcane seedlings+sugarcane stem segments and corn cob+sugarcane stem section. And the generation development period T (38.8) was short, increasing the number of generations. Therefore, Feeding the stalk borer of Chilo infuscatellus with corn cob are better than sugarcane breeding in many development indexes, and is simple and low cost.
http://hjkcxb.alljournals.net/hjkcxben/ch/reader/view_abstract.aspx?file_no=202305032&flag=1

The differences of digestive tract morphology among different developmental stages of black soldier fly were compared and the digestive tract histology in larvae was investigated. The digestive tract morphology of the 5th instar larvae, pre-pupae and adult was observed by stereoscope, while the microstructure and ultrastructure of the digestive tract (foregut, midgut and hindgut) in larvae were observed by light microscope and scanning electron microscope. The foregut (oesophagus and proventriculus), midgut and hindgut formed the digestive tract of the larvae and pre-pupae, and the length of digestive tract became shorter with larvae growth. Compared with that of larvae and pre-pupae, great changes in digestive tract morphology were observed in adult: the crop and gastric caeca appeared, while the proventriculus disappeared. The midgut length was drastically reduced and the hindgut was differentiated into ileum, colon and rectum. The results of the digestive system histology in larvae showed, the salivary gland was composed of tubular gland and ducts, which opened into the mouth. The oesophagus was composed of keratinized intimal layer and well developed muscle layers, with its end extending downward into the proventriculus. The proventriculus was a multi-layer sack with three layers. The midgut could be divided into four segments according to the morphology of the epithelial cells. The hindgut had thin and folded intestinal wall, on which a large number of rod-shaped bacteria were found. The malpighian tubules with four blind tubes were located in the junction of midgut and hindgut, with clumps of microvilli on the inner wall. The variation of digestive tract morphology with developmental stages could reflect the changes in feeding and digestive physiology. Each segment of the digestive tract in larvae had its specific histological characteristics. The foregut, midgut and hindgut may undertake the functions of food reception and preliminary digestion, digestion and absorption, and reabsorption respectively. This present study provides important morphological evidence for further understanding the digestive physiology of black soldier fly.

The microscopic anatomy, ultrastructure and growth characteristics of larval salivary glands in the parasitoid wasp, Habrobracon hebetor were observed and analyzed by dissection, photomicrography and transmission electron microscope. The salivary glands of H. hebetor larvae were a pair of colorless transparent to milky white tubular glands, extending backward from the mouth along both sides of the midgut. The unilateral gland forms a large and irregular loop in the middle, and ends in the shape of a single blind tube. The length of the salivary gland duct increases linearly with the larval instar. The ultrastructural observation of salivary gland sections showed that the gland ducts were composed of two distinct types of monolayer cells. Type I cells had thicker microvilli and contained a large number of vesicles with transporting secretory granules, besides abundant endoplasmic reticulum and mitochondria. Type II cells had short microvilli, abundant endoplasmic reticulum and mitochondria.
Dissection of the salivary gland is key to identify the components of saliva from predatory bug and investigate its functions. In this work, a case study was carried out in a predatory bug, Eocanthecona furcellata. A protocol for dissecting the complete salivary gland from this bug was described. In composition with other dissection methods, the distinct aspect of this method was using the tweezers to fix the insect body, which was helpful for dissecting and collecting the salivary gland. According to its morphology, the salivary gland of E. furcellata was composed by main gland, accessory gland, hilum and duct. Main gland can be divided into anterior main gland and posterior main gland. There was a duct connected the main gland and mouthpart, and another duct connect the main gland and accessory gland, of which both ducts were connected to hilum in the main gland. The protocol reported in this study and morphology of the salivary gland of E. furcellata can be used to guide the dissection of salivary gland from other predatory bugs to extract saliva for deciphering its components and characterizing its functions.

Cryptic species A and B are two important globally intrusive cryptic species of Leptocybe invasa, which have caused serious damage to forestry production in many countries. Due to the tiny size and undistinguishable cryptic species in morphology, it makes pest control more difficult. In this study, we researched the rapid molecular identification method of cryptic species of L. invasa, basing on the species-specific COI (SS-COI) PCR method of mitochondrial DNA cytochrome C oxidase subunit I (mtDNA COI) gene sequence. According to the COI sequence, two pairs of cryptic species A and B SS-COI primers (AF/AR and BF/BR) were respectively designed. Using these primers to amplify the DNA of the same L. invasa sample, by which could not only effectively identify the cryptic species, but also mutually verify the identification accuracy. The primer identification sensitivity detection showed that both AF/AR and BF/BR had high identification sensitivity, and their effective identification concentration thresholds for DNA were 11.42 pg/μL and 28.32 pg/μL. The rapid identification method developed for the cryptic species A and B of L. invasa in this study can addresse the challenge of discriminating cryptic species in areas invaded by L. invasa. It significantly shortens identification time, reduces identification costs, and provides technical references for further exploration of the biological differences between cryptic species A and B of L. invasa, as well as their adaptability to different resistant varieties of Eucalyptus.

To clarify the diversification and function of the sensilla on the head appendages of different instar larvae of Tomicus yunnanensis (Coleoptera∶Curculionidae∶Scolytinae), we observed the ultrastructure of the larval head by using field-emission scanning electron microscopy. The main results were as follows: 1) The structure type of the larval head was hypognathous. 2) The antennae were situated at the front view of the head with one segment and five types of sensilla that were identified as sensilla basiconica 1 and sensilla twig basiconica 1~4 with 1~3 individuals. 3) The mouthpart was consist of the labrum, mandible, maxilla, and labium. There were 12 types of the sensilla, including sensilla trichodea 1~2, sensilla chaetica 1~5, sensilla basiconica 2, sensilla digitiformia, and sensilla twig basiconica 2, 4 and 5. Each type of sensilla was presented by 4~406 individuals on each section of mouthpart. 4) Sensilla trichodea and sensilla chaetica having smooth and nonporous surfaces were widespread and regarded as mechanical sensations, sensilla basiconica and sensilla twig basiconica having the surface pores occurred only on the terminal tip of the antennae, maxillary palp, and labial palp and were considered as the chemoreceptive functions, sensilla digitiformia having only one on the ventral view of every palp might sense the signals of acoustic vibrations; 5) Sensilla type, abundance and distributions on every segment of head appendages were consistent with different instar larvae, but there were significant differences in the sensilla sizes of every type. This research will provide a scientific basis for further exploration of foraging behavior about the larvae of T. yunnanensis.

http://hjkcxb.alljournals.net/hjkcxben/ch/reader/view_abstract.aspx?file_no=202404025&flag=1

The codling moth, Cydia pomonella, is a quarantine pest that threatens the health of the fruit farming industry in China. In gene editing and other functional study techniques, homozygous mutant lines are usually screened by single pair mating strategy. Consequently, it is of paramount importance to ascertain the individual genotype prior to pairing, while avoiding any potential damage to the body. In this study, the genomic DNA (gDNA) was extracted from exuviate of the final instar larvae (10 individuals) and puparium (1, 5, 10, and 15 individuals, respectively) of C. pomonella. Then PCR amplification, agarose gel electrophoresis, and PCR product sequencing was performed to evaluate the feasibility of this method for gene detection in C. pomonella. Results showed that using the Testis specific serine/threonine protein kinase 1 (TSSK1) gene as a target gene and the gDNA extracted from 10 exuviates and 5 or more pupariums as templates, a single and bright band was amplified by PCR. The PCR products were verified as target sequence by DNA sequencing. Based on the principle of efficiency and cost saving, it is recommended to use gDNA extracted from 10 final-instar of larval exuviates or 10 pupariums to non-destructive detect the genotypes of C. pomonella by sequencing. In this study, we established a non-destructive and efficient gene detection method of C. pomonella by using the exuviates of final-instar larvae and pupariums, which laid a foundation for improving the efficiency of gene function research.

In order to efficiently monitor the population dynamics, obtain the early warning information and publish population numbers in real time, object detection algorithm YOLO V5 was introduced for deep learning to identify and count the moth on the monitoring equipment in this study. Black light trapping devices were used to obtain the adult images of P. syntaractis, and two datasets with different image sizes, enhanced by means of rotation and noise enhancement were constructed. The detection performance of different training models on acquired images and the effect of different image sizes on the recognition results of datasets were compared, and accuracy, recall rate, F1 value and average accuracy were used to evaluate the differences among the models. The 〖LM〗results showed that the accuracy, recall rate and F1 value of YOLO V5s model for the identification P. syntaractis were 96.13%, 92.06% and 0.93 respectively, and the model could well recognize the original size image. The identification and counting model based on YOLO V5 algorithm can be used in the population monitoring for its high recognition accuracy.

Chinese citrus fly, Bactrocera minax (Enderlein) (Diptera: Tephritidae), is an important pest of citrus crops. In this paper, the morphology and structure of internal reproductive system of B. minax was studied on observation of optical microscope, scanning electron microscopy and paraffin section. The results showed that the internal reproductive system of female B. minax mainly consisted of ovaries, lateral oviducts, median oviducts, spermathecae, accessory glands, anterior vagina (bursa copulatrix, ventral receptacle, and signum), and posterior vagina (vagina and ovipositor). The internal reproductive system of the male consists mainly of testis, ejaculatory pump, ejaculatory duct, male accessory gland, input ejaculatory duct, output ejaculatory duct, posterior accessory gland and aedeagus. The female had a cloaca, which was located slightly behind the anterior end of the ovipositor, while the male has no cloaca. The surface of the anterior vagina of the female was covered by two pairs of muscles, and bursa copulatrix, ventral receptacle, and signum in the interior were not easy to be observed. Ejaculatory pump was a pale yellow sphere, composed of the ejaculatory apodeme (the internal skeleton of the ejaculatory pump), muscle fibers (muscles), and ejaculatory pump sac. The morphology and structure or tissue of internal reproductive system of B. minax had evolved to adapt to the mating, egg laying and movement motion mechanism of the pseudo-ovipositor.
