In order to evaluate the sublethal effects of chlorfenapyr on F0 generation and progeny (F1 generation) of Tuta absoluta and to explore the response of detoxification enzymes and protective enzymes in F0 and F1 of T. absoluta to chlorfenapyr. This study used the leaf soaking method to determine the toxicity of chlorfenapyr to T. absoluta, and the sublethal effects of LC25 exposure to chlorfenapyr on growth and development of F0 and F1 generations of T. absoluta were analyzed by bioassay and the changes on F0 generation and progeny (F1 generation) of T. absoluta of detoxification enzymes and protective enzymes in the body after LC25 treatment were determined. The results showed that the activity of chlorfenapyr to the 2nd instar larvae of T. absoluta was high with the LC50 value of 1.33 mg/L. The number of eggs laid in the sublethal pesticide concentrations LC25 chlorfenapyr significantly reduced of F0 generation. Exposure to sublethal pesticide concentrations reduced the weight of F1 pupae, significantly prolonged the pupal period and total pre-oviposition period. In terms of detoxification enzymes, there was no significant change in carboxylesterase (CarE) activity, but glutathione S-transferase activity (GSTs) and Mixed-function oxidase (MFO) were distinctly inhibited by sublethal concentrations of chlorfenapyr on F0 generation. There was no significant change in CarE activity compared with the control, but GSTs and MFO were distinctly increased by sublethal concentrations of chlorfenapyr on F1. In terms of protective enzymes, the sublethal concentration of chlorfenapyr had no significant affect on the activities of POD and SOD on F0 generation in T. absoluta, while there was inducedeffect on the activity of CAT and the three protective enzymes CAT, POD and SOD on F0 generation. In summary, chlorfenapyr at the sublethal concentrations could significantly affect the growth and development of T. absoluta and the activity of some detoxification enzymes and protective enzymes in vivo. Therefore, this study provided valuable information for promoting the rational use of chlorfenapyr for controlling T. absoluta.

The tomato leafminer (Tuta absoluta Meyrick) (Lepidoptera) has become an important agricultural pest in China. Understanding the effects of temperature and humidity on their developmental period is essential for predicting their population dynamics. In this study, we used the hermaphroditic life table method to determine the pupation rate, feathering rate and oviposition rate of T. absoluta at 10%, 25%, 50% and 75% relative humidity, and the development period, generation cycle, larval survival rate, adult survival rate and egg production of single female by each insect state of T. absoluta at the temperatures of 15℃, 20℃, 25℃, 30℃ and 35℃. The results showed that the pupation rate and oviposition rate of T. absoluta showed a trend of increasing and then decreasing with the increase of humidity, and 75% relative humidity was the optimum humidity for its development. The development period gradually shortened with the increase of temperature under different temperature conditions, the longest development period was 63.3 d at 15°C, only 23.6 d at 30°C, and the development of generations could not be completed at 35℃. Female adults reproduced fastest and laid the highest number of eggs at 25℃, with a single female laying 112 eggs. The net value-added rate and perimeter growth rate firstly increased and then decreased with the increase of temperature, indicating that both high and low temperature were not suitable for the increase of T. absoluta population, and 25°C was the optimum development temperature.

In order to clarify the changes of cold-resistant substances in larvae, pupae and adults of the Tuta absoluta under low-temperature stress, the changes in the contents of water, fat content and cold-resistant substances (proteins, alginate and sorbitol) in each insect state were measured and analysed under low-temperature stress at 4℃, with a view to clarifying the cold-resistant substances and their characteristics in different insect states of the T. absoluta. The results showed that low temperature significantly reduced the water content of larvae and pupae from 74.46% ± 1.64% to 68.66% ± 0.90% and 70.23% ± 0.65% to 66.92% ± 0.93%, respectively; the fat content of larvae was significantly reduced from 227.64 ± 6.28 μmol/mL to 186.29 ± 6.28 μmol/mL under low temperature; while the fat content of each insect state was significantly reduced from 227.64 ± 6.28 μmol/mL to 186.29 ± 6.28 μmol/mL under low temperature. The protein content of all insect forms increased significantly at low temperature, with the greatest increase in the pupal stage, from 405.03 ± 13.678 μg/mg to 554.82 ± 29.41 μg/mg, an increase of 149.79 ± 29.41 μmol/mg. Alginate and sorbitol accumulated significantly in the pupal stage, from 31.44 ± 0.48 mg/mL to 39.82 ± 1.8 mg/mL, respectively. The significant accumulation of alginate and sorbitol from 31.44 ± 0.48 mg/mL to 39.82 ± 1.07 mg/mL and from 49.41 ± 0.72 mg/mL to 52.15 ± 0.68 mg/mL,respectively during the pupal stage indicated that they were the key chilling resistance substances during the pupal stage. Therefore, different insect forms of T.absoluta responded to low temperatures, among which the pupae of T.absoluta may rely on proteins, alginate and sorbitol to overwinter in the open ground in Ningxia.

The tomato leaf miner, Tuta absoluta (Meyrick), also known as Phthorimaea absoluta, is a significant polyphagous invasive agricultural pest. It primarily infests Solanaceae crops, such as tomatoes and potatoes, and can cause tomato yield losses of 50%~80%, or even complete crop failure. Therefore, as the “Ebola virus of tomatoes”, T. absoluta was first found in Xinjiang, China, in 2017, and has broken out in several regions of China. T. absoluta often occurs covertly, and its suitable habitats are widespread in China. This pest can transmitted over long distances through the transport of tomato seedlings and fruits, indicating a clear trend of further spread. Therefore, there is an urgent need to develop efficient and low-cost detection and identification technologies and methods for T. absoluta, in order to achieve early detection and advance warning of this significant invasive species.Currently, the primary techniques used to identify T. absoluta include traditional morphological identification methods, molecular biological detection techniques, and artificial intelligence-based image recognition methods. In this paper, the research status of detection and identification technology of T. absoluta was reviewed, and its development trend was prospected, aiming at providing technical support for delaying the spread and diffusion of T. absoluta and targeting strategies.

http://hjkcxb.alljournals.net/hjkcxben/ch/reader/view_abstract.aspx?file_no=202501003&flag=1

Tuta absoluta is a globally invasive pest originating from South America that primarily damages Solanaceous crops. Currently, the main control method for T. absoluta is chemical control, but chemical control can lead to an increase in pest resistance, causing damage to the environment, non target species, and even negative impacts on human health. Therefore, environmentally friendly biological control techniques have become a research focus and have gradually replaced chemical control in several countries and regions for managing T. absoluta. Entomopathogenic nematodes (EPNs) are commonly used as biological control agents in pest management and are an important part of Integrated Pest Management (IPM) strategies. At present, the research on biological control of T. absoluta using EPNs is relatively scattered. This paper systematically reviews the research progress of using EPNs to control T. absoluta globally, and summarizes it from the following parts: (1) classification and biological characteristics of EPNs; (2) research progress on using EPNs to control T. absoluta; (3) effects of environmental factors on the efficacy of EPNs; (4) combined use of EPNs with other control measures. It summarizes the methods for enhancing the controlling efficacy of EPNs on T. absoluta and provides prospects for future research on using EPNs for the management of T. absoluta.

From June to July 2024, during greenhouse in Linzhi City (Tsatsum Town) and filed surveillance in Changdu City (Karuo Town and Rumei Town), Xizang, China, we successively found serious infestations of lepidopteran larvae on tomato and potato plants, with mined leaves and attacked fruits. The lepidopteran was identified as Tuta absoluta (Meyrick) using morphology and molecular methods, a major agricultural invasive pest in China. It is the first report that T. absoluta occurred in Xizang, China. In this paper, the morphological characteristics of T. absoluta and its infestation characteristics in Xizang are described. The possibility that the South American tomato leafminer could be introduced into Xizang via India through international trade in agricultural products, or from some domestic regions where the pest has already occurred (Sichuan, Yunnan, etc.) through seedling transportation was discussed and the spread trend was studied and judged. Currently, it is evident that the diffusion trend of T. absoluta is from low-altitude to high-altitude in Xizang. Therefore, in order to prevent the continued introduction and diffusion hazard of the pest, it is necessary to strengthen the inspection and quarantine on agricultural product trade and seedling transportation to block dispersion pathways of T. absoluta, and timely and comprehensive survey of the pest in the entire region, including clarifying its main occurrence regularity, degree of damage, and distribution range and so on, which provides reference for monitoring and controlling agricultural invasive species in Xizang.

In order to develop a better control strategy for Tuta absoluta on different solanaceous vegetables, the population dynamics of T. absoluta on tomato, eggplant and pepper under two cultivation methods (open field and protected greenhouse) were investigated in Kukelanmu vegetable base of Kaerdun Township, Yining County, Yili Kazak Autonomous Prefecture for two consecutive years from 2023 to 2024, and the population dynamics of T. absoluta on three common solanaceous vegetables were compared. At the same time, the damage degree of T. absoluta to tomato, eggplant and pepper leaves was evaluated by the five-degree evaluation method of plant damage. The results showed that the damage degree of T. absoluta to tomato leaves was the highest. The number of leaves with a single leaf damage area more than 35% (V level) reached as high as 45.6% of the total number of leaves in the plant, and the average fruit-boring rate of tomato fruit reached as high as 7.8%. The damage degree of T. absoluta to eggplant leaves was relatively low, and the number of leaves with a single leaf damage area more than 35% (V level) reached as high as 4.2% of the total number of leaves, and no damage to eggplant fruit was found. T. absoluta was not found to damage pepper leaves and fruits. The maximum amount of adult, egg, larva and pupa of T. absoluta on tomato and eggplant was from June to August, and the number of these four stages on tomato was more than that on eggplant. The field investigation did not find the damage symptoms of T. absoluta on pepper, and the eggs, larvae and pupae of T. absoluta had not been detected. However, a large number of adults could be trapped in the pepper fields of open field and protected greenhouse. This could be due to the presence of Solanaceae weeds such as Solanum nigrum in the pepper field. On the other hand, it could be that tomatoes and eggplants were not planted around the pepper field, and the planting distance was less than the attraction distance of the sex lure (20 m). In summary, the T. absoluta in Ili area mainly appeared from June to August, and among the three vegetables, it mainly harmed tomato, followed by eggplant, and did not harm pepper at all. Different control strategies should be adopted. This study is helpful to detect and control the T. absoluta in time, reduce economic losses, and provide a theoretical basis for the development of comprehensive prevention and control technology for the major invasive pest T. absoluta.

Glutathione-S-transferases (GSTs) play an important role in insect adaptation to plant secondary metabolites. To clarify the role of the GST gene in the detoxification of Tuta absoluta to α-tomatine, we first cloned the full-length sequence of TabsGSTs2 gene. Bioinformatic analysis was employed to analyze the sequence characteristics, physicochemical properties, conserved domains, gene structure, and evolutionary relationships of the TabsGSTs2 gene. The expression levels of TabsGSTs2 under α-tomatine stress were measured using RT-qPCR, and the binding capacity and mode of TabsGSTs2 to α-tomatine were investigated using homology modeling and molecular docking. The results showed that the CDS of TabsGSTs2 was 609 bp, encoding 203 amino acids, with a theoretical isoelectric point of 8.47 and a molecular weight of 23.895 kDa. TabsGSTs2 contains four β-sheets and nine α-helices, and had the typical conserved domains of GST genes, including the GSH binding site (G-site) and the substrate binding site (H-site). Phylogenetic analysis indicated that TabsGSTs2 belongs to the sigma subfamily and was closely related to CpGSTs2 of Cydia pomonella. Under α-tomatine stress, the expression level of TabsGSTs2 was significantly higher than that of the control at 72 h. Molecular docking results suggested that TabsGSTs2 bind strongly to α-tomatine, with the interaction being stabilized primarily by hydrogen bonds, hydrophobic forces, and salt bridges. Our study provides reference for subsequent studies of the molecular mechanisms of T. absoluta adaptation to α-tomatine and provides a basis for further exploration of new targets for the control of T. absoluta.

Insect gustatory receptors are a crucial class of chemoreceptors that can detect external environmental stimuli and influence insect behaviors such as host selection, feeding, mating, and oviposition. This study focused on the invasive pest Tuta absoluta (Meyrick) that affected tomatoes. To elucidate the characteristics of the gustatory receptor (GR) gene family in T. absoluta and investigate the expression patterns of GR genes across different developmental stages, we employed bioinformatics approaches to identify, chromosomally locate, conduct phylogenetic analysis, and analyze age-stage expression profiles of the GR genes in T. absoluta. The results indicated that 62 GR genes of the T. absoluta moth were identified and distributed on 19 chromosomes. The length of the encoded protein sequence ranged from 318 to 2034 bp, with an average of 5 exon fragments and 1 to 7 transmembrane regions. Phylogenetic analysis revealed that the GR genes of the T. absoluta could be categorized into four distinct subfamilies: bitter taste receptors (35 genes), carbon dioxide receptors (1 gene), sugar receptors (5 genes), and Gr43a-like receptors (7 genes). The remaining genes have not been classified yet. Expression analysis across different developmental stages revealed that the GR genes of the T. absoluta exhibited significant variation in expression levels throughout its life cycle. The research results provided a solid foundation for further elucidating the functions of gustatory receptor genes in the T. absoluta and uncovering its co-evolutionary relationship with host plants. These findings offered scientific evidence for controlling the T. absoluta by functional plants and lay the foundation for developing novel pest control technologies from a gustatory receptor perspective.

The gut bacteria composition in phytophagous insects is closely linked to their developmental stages and can fulfill multiple functions for the host insect. However, few studies have investigated the relationship between gut bacteria and developmental stages in the tomato leaf miner moth, Tuta absoluta (Meyrick). This study aimed to characterize the diversity of gut bacteria in larvae of different instar stages of T. absoluta and to predict the functional roles of these bacteria. We extracted genomic DNA from the gut bacteria of T. absoluta larvae at the 1st, 2nd, 3rd, and 4th instar stages, and analyzed the diversity and abundance of the gut bacteria using Illumina MiSeq II high-throughput sequencing technology. Alpha diversity analysis indicated that gut bacterial diversity and abundance peaked in the 1st instar larvae of T. absoluta and diminished as instar age increased. The dominant phylum of gut bacteria across all four instar stages was Proteobacteria. Excepting that Enterobacteria was the dominant genus in the 4th instar larvae, whereas Wolbachia was predominant in the other instars. Predicted functional analysis based on the PICRUSt2 gene database revealed that the gut bacteria in T. absoluta larvae participated in carbohydrate, amino acid, and energy metabolism, suggesting their crucial roles in digestion, absorption, and nutrient provision. Among them, there were significant differences in the metabolism fuction of intestinal bacteria at different instars and the 1st instar larvae were significantly higher than that in the 3rd and 4th instars. This study provides a foundational basis for exploring the interactions between gut bacteria and their host in T. absoluta and offers additional insights and directions towards the adaptive evolution and integrated management of this pest.

http://hjkcxb.alljournals.net/hjkcxben/ch/reader/view_abstract.aspx?file_no=202501010&flag=1

The tomato leaf miner, Tuta absoluta Meyrick (Lepidoptera), primarily attacks Solanaceae species, particularly tomatoes, significantly affecting both yield and quality. Saliva plays a crucial role in insect-plant interactions which needs direct physical contact. As an important part of insect saliva, it is of significant importance to analyze the role of salivary bacteria in the feeding processes of insects on host plants. In this study, conventional isolation methods were employed to isolate bacteria from the saliva of third-instar larvae of T. absoluta. Species identifications of the isolated strains were performed using 16S rRNA sequence homology analysis. The isolated strains were inoculated into tomato plants, and growth and defense indexes, including chlorophyll, malondialdehyde, peroxidase, superoxide dismutase, soluble sugar, soluble protein, nitrogen, and phosphorus were measured after inoculation. The results showed that the isolated salivary bacteria belonged to three phyla, six genera, and seven species: Enterococcus mundtii and Bacillus amyloliquefaciens (phylum Firmicutes), Enterobacter cloacae and Acinetobacter rhizosphaerae (phylum Proteobacteria), Microbacterium arborescens, Microbacterium oleivorans and Curtobacterium flaccumfaciens (phylum Acinobacteria). Inoculation experiments in vitro demonstrated that M. arborescens and E. mundtii significantly reduced the peroxidase activity of tomato plants which decreased by 62.92% and 76.83% after treatment, respectively. Treatments with A. rhizosphaerae and E. mundtii resulted in a significant increase in malondialdehyde content in the plants (P < 0.05). Soluble protein levels increased significantly after treatment with M. arborescens, E. mundtii, and M. oleivorans (P < 0.05), whereas soluble sugar content decreased significantly after treatment with A. rhizosphaerae and E. cloacae (P < 0.05). Nitrogen levels also exhibited a decreasing trend after treatment with the salivary bacteria. Additionally, chlorophyll content was significantly higher in all treatment groups than that of the control group (P < 0.05). In conclusion, inoculation in vitro with salivary bacteria from T. absoluta leads to peroxidation in tomato plants, and certain strains inhibit the plants' defense mechanisms. This finding indicates that the saliva of T. absoluta plays a role in regulating the growth and defense responses of tomato plants, which may be a factor of facilitating the adaptation of T. absoluta to tomato. This study enriches our understanding of the mechanisms underlying the interaction between T. absoluta and tomato plants and provides a theoretical basis for the prevetion and control of T. absoluta.

The physicochemical trapping and control technology is a method for the prevention and control of crop pests and diseases by means of physicochemical measures, including the utilization of pheromones, lights, sticky traps, etc. To clarify the performance effect of the sex pheromone product of Tuta absoluta in the field, four kinds of sex pheromone lures (Y-ZJ, Y-ZD, Y-NK, Y-ZZ) (table 1) and three kinds of sex pheromone diffusers (M-ZJ, M-CBT, M-NK) (table 2) were selected. These products were developed by various enterprises and research institutions to assess the efficacy of the technology against T. absoluta in field trials conducted in Yining City, Yili Region, Xinjiang Province. The scores of the sex pheromone lures were based on the maximum trapping volume, average daily trapping volume, duration of efficacy, and unit price. The scores, in descending order, were Y-ZD, Y-ZZ, Y-ZJ, Y-NK, and Y-CK. After 60 days, Y-ZD exhibited the highest larvae control efficiency (23.54%), leaf damage control efficiency (4.33%), and fruit damage control efficiency (14.81%). The average mating disruption rates for the three products were as follows: M-NK (93.68%), M-ZJ (93.54%), and M-CBT (86.13%). Similarly, the average larvae control efficiencies were M-NK (85.27%), M-ZJ (80.90%), and M-CBT (66.43%), while the average leaf damage control efficiencies were M-NK (88.74%), M-ZJ (79.17%), and M-CBT (60.82%). The results showed that the efficacy of using sex pheromone lures in the field was low, and they could only be used for monitoring. However, Y-ZD demonstrated relatively higher efficacy. The average mating disruption rate of the three sex pheromone diffusers exceeded 85%, while the average larvae control efficiency and leaf damage control efficiency exceeded 65% and 60%, respectively. Among them, M-NK demonstrated the best mating disruption effect and was recommended for field application, which was at a control cost of 125 yuan per mu (approximately 0.0667 hectares).
